摘 要:为明确从茶园腐殖层昆虫尸体上分离的虫生真菌的分类地位及其对茶尺蠖的生防潜力,采用形态学特征观察和rDNA−ITS序列分析方法对该菌株进行鉴定,并测定该菌株对茶尺蠖4龄幼虫的毒力及其胞外酶活性。结果表明,从茶园腐殖层虫尸上分离获得一株金龟子绿僵菌CHMA−005;该菌株侵染茶尺蠖4龄幼虫10 d后,致死率、LC_(50)和LT_(50)分别可达100%、1.84×10^(4) cfu/mL和3.57 d;蛋白酶活性和几丁质酶活性均在第6 d达到最大值,分别为31.43和27.73 U/mL。可见,金龟子绿僵菌CHMA−005对茶尺蠖的致病力强,胞外酶活性高,具有良好的生防潜力和应用前景。Fungal strains were identified using morphological characteristics and rDNA-ITS sequence from entomogenous fungi isolated from insect cadaver in the humic layer of tea garden and evaluated for their biocontrol potential and extracellular enzyme activities against the 4th instar larvae of Ectropis obliquus.The isolated fungus was identified as Metarhizium anisopliae CHMA-005.After infection of the 4th instar larvae of E.obliquus for 10 days,the strain CHMA-005 resulted in mortality,LC_(50) and LT_(50) of 100%,1.84×10^(4) cfu/mL and 3.57 d,respectively.Protease activity and chitinase activity peaked on the 6th day after infection,which were 31.43 U/mL and 27.73 U/mL,respectively.Therefore,with high pathogenicity and extracellular enzyme activities to E.obliqua,M.anisopliae strain CHMA-005 has good biocontrol potential and application prospect.
