摘 要:目的:研究生、熟大黄对大承气汤泻下作用的影响。方法:将72只小鼠随机分为空白对照组、阳性对照组及生、熟大黄大承气汤低(6 g/kg)、高(10 g/kg)剂量组,共6组,每组12只。分别用代谢笼积分法、炭末推进作用及Na^(+)-K^(+)-ATPasc活性实验,比较各组泻下作用、炭末推进率及Na^(+)-K^(+)-ATPasc活性的差异。结果:生大黄大承气汤组、熟大黄大承气汤组与空白对照组及阳性对照组比较,其对正常小鼠泻下作用、炭末推进作用、抑制Na^(+)-K^(+)-ATPase活性的作用差异均有统计学意义(P<0.05),且生大黄大承气汤各组与熟大黄同剂量组比较泻下作用更显著(P<0.05),并与给药剂量成正相关。结论:生、熟大黄对大承气汤泻下作用的影响差异有统计学意义,这可能与君药生、熟大黄的主要化学成分含量变化引起大承气汤复方中蒽醌类化合物的溶出量变化有关。Objective:To study the impacts of raw and prepared rhubarb on laxative effects of Dachengqi decoction.Methods:All 72 mice were randomized into blank control group,positive control group,low(6g/kg)and high(10g/kg)doses groups of raw and prepared rhubarb Dachengqi decoction,six groups altogether,12 mice each group.To compare the difference in laxative effects,advance rate of carbon powder and Na^(+)-K^(+)-ATPasc activity in different groups using metabolic cage integral method,carbon powder propulsion and Na^(+)-K^(+)-ATPasc activity experiment respectively.Results:When raw rhubarb Dachengqi decoction group and prepared rhubarb Dadnenggi decoction group were compared with blank control group and positive control group,the differences existed in their effects on laxative effects,advance rate of carbon powder and inhibiting Na^(+)-K^(+)-ATPasc activity(P<0.05),laxative effects were more notable when the groups of raw rhubarb Dachengqi decoction were compared with equivalent dose groups of prepared rhubarb(P<0.05),and it was positively related to medication dose.Conclusion:The impacts of raw and prepared rhubarb are different on laxative effects of Dachengqi decoction,and this might be related to the changes in the dissolution of anthraqui-nones of Dachengqi decoction induced by the changes of main chemical ingredients in monarch raw and prepared rhubarb.
