摘 要:目的:探讨双氢杨梅树皮素(ampelopsis,APS)通过群体效应(quorum sensing,QS)对体内金黄色葡萄球菌(staphylococcus aureus,SA)的抗菌作用。方法:对标准SA NTCC8325及其QS调控基因缺失株NTCC8325ΔagrA、NTCC8325luxS进行生长曲线测定,以确定3个株菌的活力最强时期;检测高、中、低剂量APS给药后3个株菌腹腔感染小鼠腹腔液活菌数和小鼠血液白细胞数。结果:标准SANTCC8325活力最强时期为10~12 h,NTCC8325ΔagrA、NTCC8325ΔluxS活力最强时期为12~14 h;APS对3个株菌腹腔感染小鼠均有保护作用,且在一定范围内呈量效关系;APS对NTCC8325ΔluxS腹腔感染小鼠的保护作用较APS对NTCC8325ΔagrA腹腔感染小鼠的保护作用强。结论:APS在体内通过直接影响agr QS系统及间接影响lux QS系统,从而产生抗SA的作用。Objective:To explore antibacterial effects of APS on SA in vivo based on QS.Methods:The growth curves of standard SA NTCC8325 and its QS regulatory gene delection strains NTCC8325ΔagrA and NTCC8325luxS were determined,to confirm the most vigorous period of three strains;to measure the number of viable bacteria in peritoneal fluid of mice with abdominal infection and the number of white blood cells after administering with high,moderate and low dosages of APS.Results:The most vigorous period of standard SA NTCC8325 was from 10 to 12 hours,the periods of NTCC8325ΔagrA and NTCC8325luxS from 12 to 14 hours;APS owns protective effects on the mice suffering from three strains of bacteria infection in abdominal cavity,and it presented the dose-dependent relationship within a certain range;protective effects of APS on the mice with abdominal infection of NTCC8325ΔluxS were stronger compared with its effects on the mice with abdominal infection of NTCC8325ΔagrA.Conclusion:APS could produce anti-SA effects through directly affecting agr QS system and lux QS system indirectly in vivo.
