摘 要:目的:基于网络药理学理念,探讨“黄芪-当归”治疗慢性再生障碍性贫血(CAA)的作用机制。方法:利用中药系统药理学分析平台(TCMSP)与Pubchem数据库获取黄芪、当归的活性成分及其平面结构图与结构式,通过SwissTargetPrediction服务器和人类基因数据库(GeneCards)获取该药对治疗CAA的成分-疾病靶标及其Uniprot ID。根据成分-疾病靶标与活性成分的作用关系,筛选出有效成分,根据成分-疾病靶标之间的相互作用关系筛选出5个核心靶标,并利用systemsDock在线工具进行分子对接验证。通过DAVID6.8在线工具进行成分-疾病靶标的基因本体论(GO)分析与基因组百科全书(KEGG)通路富集分析。结果:筛选出黄芪-当归活性成分共20个,可作用于144个靶标,主要是调控PI3K-Akt信号通路和VEGF信号通路。结论:“黄芪-当归”可能是通过改善造血微环境、促进红细胞生成、增加血管新生等多方面调节造血功能治疗CAA。Objective:The study aims to explore the the active components of"astragalus-angelica"("As-An")and its possible mechanism in the treatment of chronic aplastic anemia(CAA)based on the network pharmacology.Method:Active components in"As-An"were researched in the traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP).The targets of"As-An"treating CAA and their Uniprot ID were screened by using SwissTargetPrediction and GeneCards databases.The effective components were found according to the relationships between active components and the potential targets.The potential targets were get from the Protein-protein interactions(PPI).Molecular docking between the potential targets and the effective components of"As-An"was performed by Systems Dock Web Site.Gene ontology(GO)enrichment analysis and Kyoto encyclopedia of genes and genomes(KEGG)pathway analysis of the targets were also carried out to explore the mechanism of"As-An".Result:20 effective components reaching the standard were gathered and they act on 144 targets.5 potential targets were found.GO and KEGG analysis suggest that"As-An"may impact the PI3K-Akt signaling pathway,VEGF signaling pathway to relieve the process of CAA.Conclusion:This study proved that themechanism of"As-An"for CAA may include improving hematopoietic microenvironment,promoting erythropoiesis,increasing angiogenesis,and so on.
