摘 要:目的明确S期激酶相关蛋白2(S-phase kinase associated protein 2,Skp2)在食管鳞癌(esophageal squamous cell carcinoma,ESCC)细胞中的功能。方法利用RT-qPCR和Western blot法检测Skp2基因在ESCC细胞系中的表达;用慢病毒敲低Skp2基因在ESCC细胞系中的表达后,采用MTT法、克隆形成试验、Transwell小室试验检测Skp2对ESCC细胞增殖、迁移的影响,采用RT-qPCR法检测Skp2潜在下游靶基因P53、P300的表达。结果与正常食管上皮细胞相比,Skp2基因在ESCC细胞系中呈现高表达的趋势。利用慢病毒敲低Skp2基因的表达可明显抑制ESCC细胞系的增殖及迁移能力;Skp2下游潜在靶基因P53、P300表达量显著上升。结论 Skp2基因可通过抑制P53、P300等抑癌基因的表达,促进ESCC细胞的增殖、迁移及克隆形成。Objective To clarify the role of S-phase kinase associated protein 2(Skp2) in esophageal squamous cell carcinoma(ESCC). Methods The expression of Skp2 gene in ESCC cell lines was determined by RT-PCR and Western blot. After the expression of Skp2 in ESCC cell lines was knocked down by lentivirus,the effects of Skp2 on proliferation and migration of ESCC cells were evaluated by MTT assay,colon formation test and Tanswell assay. The expressions of potential downstream genes P53 and P300 of Skp2 were determined by RT-qPCR. Results Compared with that in normal esophageal epithelial cells,the Skp2 gene showed a trend of high expression in ESCC cells. The knocking down of expression of Skp2 gene by lentivirus significantly inhibited the proliferation and migration of ESCC cell lines,while increased the expression levels of potential downstream genes P53 and P300 significantly. Conclusion Skp2 gene may promote the proliferation,migration and clone formation of ESCC cells by inhibiting the expression of tumor suppressor genes such as P53 and P300.
