摘  要:用60Coγ射线辐照诱变粳稻‘嘉花1号’,得到一个能够稳定遗传的低温白化致死突变体tcd34,并对其进行表型鉴定与基因定位研究。在低温20℃下,与野生型‘嘉花1号’相比,突变体tcd34在整个苗期均表现为完全白化,光合色素含量明显降低,且叶绿体结构不完整,植株最终死亡;在高温32℃下,该突变体苗期叶片叶色、光合色素含量及叶绿体发育基本恢复正常。利用tcd34和培矮64S杂交得到的F2代群体,将突变基因定位于3号染色体In Del分子标记ID03M14453和ID03M14723之间的341 kb内,包含15个候选基因。通过转录水平的分析,发现在低温20℃下,该突变体中参与叶绿素合成及叶绿体发育相关基因(CAO,Cab1R,FtsZ)的表达水平有不同程度的下调,说明该突变基因会影响低温环境下水稻的正常生长。The rice thermo-sensitive albino phenotype mutant,tcd34,was obtained from japonica rice variety’Jiahua 1’(wild-type,WT)irradiated with60 Co gamma-ray,and its phenotype identification and genetic analysis were carried out.compared with WT,the tcd34 mutant showed albino phenotype throughout all seedling stage,with a significant decrease in chlorophyll content and incomplete chloroplast structure,and died eventually.However,at high temperature of 32℃,leaf color,photosynthetic pigment content and chloroplast development of the mutant at seedling stage were basically restored to normal.Then using the Pei’ai 64 S/tcd34 F2 population,TCD34 gene was located in 341 kb region between ID03 M14453 and ID03 M14723 makers on chromosome 3,which spanned four BACs and contained 15 candidate genes.Additionally,the transcription levels of three genes related to chlorophyll synthesis and chloroplast development(CAO,Cab1 R,FtsZ)were significantly down-regulated at 20℃,showing that TCD34 plays an important role and can affect the growth of rice at low temperatures.
