摘  要:肉桂酰辅酶A还原酶(cinnamoyl-CoA reductase,CCR)在木质素生物合成途径中起着关键作用。本研究依据转录组数据设计特异性引物,采用RT-PCR方法从竹叶花椒(Zanthoxylum armatum)中克隆得到一个全新的CCR基因的全长c DNA序列,命名为ZaCCR。序列分析结果表明,ZaCCR包含完整的cDNA开放阅读框(OFR),由990 bp组成,编码329个氨基酸。Blast比对结果显示该蛋白质属于CCR家族蛋白;系统进化树结果显示竹叶花椒(Zanthoxylum armatum)与芸香科植物甜橙(Sweet orange,Citrus sinensis)、蜜柑(Satsuma mandarin,Citrus unshiu)等亲缘关系较近。荧光定量PCR检测显示,ZaCCR在竹叶花椒中的表达量从大到小分别为:嫁接树的茎、实生树的茎、嫁接树的叶、实生树的叶。Cinamoyl-CoA reductase plays a key role in lignin biosynthesis pathway.In this study,we designed specific primers based on transcriptome data and cloned a novel full-length cDNA sequence of CCR gene from Zanthoxylum armatum by RT-PCR,named ZaCCR.Sequence analysis showed that ZaCCR contains a complete open reading frame(ORF),consisting of 990 bp,encoding 329 amino acids.Blast comparison results showed that the protein belongs to CCR family,and phylogenetic tree results showed that Zanthoxylum armatum was closely related to Rutaceae plants such as sweet orange(Citrus sinensis)and satsuma mandarin(Citrus unshiu).Fluorescence quantitative PCR showed that the expression of Za CCR in Zanthoxylum armatum from large to small are:Stem of grafted tree,stem of seedling tree,leaf of grafted tree and leaf of seedling tree.
