摘 要:本研究结合土壤理化性质,采用Illumina MiSeq高通量测序技术对莲雾根际土壤细菌群落进行16S rRNA V3~V4区检测,以分析不同类型催花肥对莲雾根际土壤细菌群落结构的变化,并探索土壤理化性质与土壤细菌群落多样性之间的相关性。结果显示:(1)共检测到7688个操作分类单元(OTU),包含167736条有效序列,可分为28个门418个属。(2)相对丰度和多样性分析显示,不同类型催花肥处理的细菌群落多样性及丰度顺序为BSH(香蕉茎秆有机肥处理)>BMH(香蕉茎秆有机菌肥处理)>CK(对照)>IFH(无机肥处理)>GMH(羊粪有机肥处理)。其中,由酸杆菌、绿弯菌、变形菌和放线菌所组成的优势菌群占总群落的85.16%。(3)分析环境因子的相关性热图可知,pH值、w(TN)、w(速效磷)、w(速效钾)和w(有机质)对WCHB1-60、SM2F11、装甲菌门、放线菌门、厚壁菌门、变形菌门和硝化螺旋菌门7类菌群的影响较为显著。因此,在有机肥中添加一定量的香蕉茎秆废弃物,不仅能够提高土壤细菌群落的多样性及丰度,维持土壤生态微环境,还有利于香蕉茎秆废弃物的可持续利用。This study considering the soil physical and chemical properties,the bacterial community in the rhizosphere soil of wax apple in 16S rRNA V3~V4 region was detected by the Illumina MiSeq high-throughput sequencing technology so that it was possible to analyze the bacterial community structure of rhizosphere soil in different types of flower forcing fertilizers.Furthermore,based on the result,to explore the correlation between soil physical and chemical properties and soil bacterial community diversity.As a result,it showed that(1)7688 OTU(Operational taxonomic unit)were detected in all samples,including 167736 effective sequences,which could be divided into 28 phyla and 418 genera.(2)The relative abundance and diversity index analysis showed that the order of diversity and abundance of bacterial community in different types of fertilizer treatment was as follows:BSH(Banana stalk organic fertilizer)>BMH(Banana stalk organic bacterial fertilizer)>CK(Ccontrol group)>IFH(Inorganic fertilizer)>GMH(Sheep manure organic fertilizer treatment).Above all,Acidobacteria,Chloroflexi,Proteobacteria and Actinobacteria were the dominant bacteria,which accounting for 85.16%.(3)Correlation thermal imagine analysis for environmental factors showed the effect of the environmental factors such as pH,TN,Available P,Available K and Organic Matter impacted on WCHB1-60,SM2F11,Armatimonadetes,Actinobacteria,Firmicutes,Proteobacteria and Nitrospira were more significant.Therefore,adding the waste of banana stalk to organic fertilizer can not only improve the diversity and abundance of soil bacterial community as well as maintain the soil ecological microenvironment,but also facilitate the sustainable use of the waste of banana stalk.
