摘 要:为进一步了解猪伪狂犬病病毒新流行株的变异特点,2018年对来自河南省济源市某猪场疑似猪伪狂犬病的病料组织进行PRV的分离鉴定,并分析其生物学特性。结果表明,分离株HNJY对小鼠具有致病性;HNJY株gE、gB、gD与国内毒株的核苷酸和氨基酸同源性分别高达99.2%~99.9%和98.6%~100%、99.7%~100.0%和99.2%~100.0%、99.4%~99.9%和99.3%~99.8%,gC基因与2011年之后分离的国内变异株的核苷酸和氨基酸同源性分别高达99.8%~100.0%和99.4%~100.0%,TK基因与2011年之后分离的国内变异株的序列同源性高达100%。从遗传进化来看,gE、gB基因可作为鉴别中国毒株和欧美毒株的标记基因,gC基因与分离年代有一定联系,gD、TK基因与分离年代无明显关联。综上表明,分离株HNJY株为新的国内变异株。In order to understand the variation characteristics of PRV,we isolated and identified PRV from pig tissue from a pig farm suspected with pseudorabies disease in Jiyuan city,Henan province in 2018,and then analyzed its biological characteristics.The results showed that the HNJY strain was pathogenic to mice.The nucleotide and amino acid homology of the HNJY strain gE,gB,and gD with domestic strains were as high as 99.2%~99.9%and 98.6%~100%,99.7%~100.0%and 99.2%~100.0%,99.4%~99.9%and 99.3%~99.8%.The nucleotide and amino acid homology of the gC gene of the HNJY strain with domestic mutant strains isolated after 2011 were as high as 99.8%~100.0%and 99.4%~100.0%,respectively.The sequence homology of TK gene of the HNJY strain and domestic mutant strains isolated after 2011 as high as 100%.From the perspective of branching and genetic distance,gE and gB genes can be used as marker genes to identify Chinese strains and European and American strains.The gC gene had a certain relationship with the age of separation,and the gD and TK genes had no obvious correlation with the age of separation.In summary,the isolated HNJY strain is a new domestic mutant strain.
