摘  要:目的:探讨外涂黄连素溶液对脊髓损伤(SCI)SD大鼠撕咬伤口行为的干预效果。方法:将12只SD雌鼠随机分为黄连素涂抹组与生理盐水涂抹组,采用改良Allen法自制打击装置造成大鼠的T10段脊髓急性损伤,黄连素涂抹组给予0.05%黄连素生理盐水溶液每天涂抹背部伤口2次,生理盐水组给予生理盐水涂抹相同部位,观察两组SD大鼠术后7 d内背部伤口撕咬的发生率,用Basso-Beattie-Bresnahan(BBB)法判断SD大鼠运动恢复情况,酶联免疫吸附实验(ELISA)检测血清炎症因子白介素-1β(IL-1β)和肿瘤坏死因子-α(TNF-α)的变化,使用苏木精-伊红(HE)染色和尼氏染色分别评估脊髓的空洞率及神经元细胞数量变化情况。结果:黄连素涂抹组的SD大鼠发生背部伤口撕咬次数3次(7.1%),低于对照组13次(30.1%),对血清炎症因子IL-1β和TNF-α的表达没有影响(P<0.05),对脊髓组织损伤空洞率及神经元细胞数量没有影响(P<0.05)。结论:外涂黄连素溶液能有效减少SCISD大鼠伤口的撕咬行为。Objective: To investigate intervention effect of topically berberine solution on the bite wound in SD spinal cord injured(SCI)rats. Methods: A total of 12 SD rats were randomly divided into spinal cord injury group, 0.05 % berberine group and normal saline smear group. Acute spinal cord injury model of T10 spinal cord in rats were established by using the modified Allen’s method, to observe the incidence of wound secondary bite,changes of serum inflammatory factors and spinal cord repair in SD rats. For the berberine group, 0.05% berberine solution was applied to the wound twice a day. For the saline group, the same site was applied with saline.The incidence rates of the back wound in two groups with 7 days were record. The motor function of SCI rats in two groups were evaluated by Basso-Beattie-Bresnahan(BBB)score. The serum inflammation cytokines of interleukin-1β(IL-1β)and tumor necrosis factor-α(TNF-α)were detected by enzyme-linked immunosorbent assay(ELISA). The ratio of traumatic syringomyelia and the number of Nissl positive cells were evaluated by the H&E andNisslstaininginspinal tissue. Results: The rate of back wound was three times(7.1%)in SD rats treated with0.05% berberine saline solution was lower than the rate of back wound in the control group thirteen times(30.1%), and the expression of serum inflammatory cytokines IL-1β and TNF-α were not difference(P<0.05)and had no effect on the ratio of traumatic syringomyelia and the number of Nissl positive cells in spinal cord tissue(P<0.05).Conclusion: Applying the berberine solution to wound can effectively reduce the biting behavior of SD rats with spinal cord injury.
