摘　　要：目的:研究铁死亡诱导剂RSL-3对造血干/祖细胞在体外分化成巨核细胞中的作用。方法:收集广西医科大学第一附属医院住院的3例健康供者的骨髓采集物,利用磁珠分选出CD34+细胞(造血干/祖细胞),添加无血清培养基及巨核细胞定向扩增因子培养14 d,将RSL-3处理的细胞作为实验组,相同溶剂浓度的DMSO处理的作为对照组,处理96 h后通过流式细胞术观察两组CD41+细胞含量、脂质过氧化水平,并进行倍性分析。结果:实验组CD41+细胞含量以及脂质过氧化水平均较对照组明显降低(P<0.05)。倍性分析显示,两组巨核细胞成熟情况无明显差异(P<0.05)。结论:铁死亡诱导剂RSL-3可以抑制造血干/祖细胞向巨核细胞分化,可能与细胞内氧化还原状态的改变有关。Objective: To investigate the effect of RSL-3, a ferroptosis inducer, on differentiation of hematopoietic stem/progenitor(CD34+cells) into megakaryocytes. Methods: The bone marrow samples of 3 healthy donors were collected, then the CD34+cells were purified using a magnetic human CD34 MicroBead Kit. CD34+cells were cultured in serum-free expansion medium supplemented with cytokines which selectively promoted the expansion and differentiation of megakaryocytes for 14 days. The cells were treated with DMSO(control group) or RSL-3(experimental group) for 96 hours. The percentage of CD41+cells, lipid peroxidation and ploidy analysis were detected by flow cytometry. Results: Compared with the control group, the percentage of CD41+cells and lipid peroxidation level were decreased in the experimental group(P<0.05), whereas nosignificant difference was found between the two groups in maturation of megakaryocytes by ploidy analysis(P>0.05). Conclusion:Ferroptosis inducer agent RSL-3 inhibited CD34+cells differentiation into megakaryocytes, and this effect may be dependent on the change of redox environment.
