摘  要:目的了解引起2020年10月大连市西岗区一起儿童急性肠胃炎疫情的诺如病毒基因亚型,为该类疾病的预防和控制提供参考数据。方法提取患儿粪便标本中的病毒RNA,应用逆转录聚合酶链反应(reverse transcription-polymerase chain reaction,RT-PCR)进行诺如病毒特异性核酸片段检测并进行测序,使用Lasergen 7.1、Norovirus Typing Tool Version 2.0、Clustal X 2.1和MEGA 10.1等软件对毒株的基因序列片段构建Neighor-joining系统进化树并进行种系进化分析。结果 2020年10月大连市西岗区一起儿童急性肠胃炎疫情的11份患儿标本中检出NV GⅡ型核酸阳性,其中1份经RT-PCR扩增并测序后,构建系统进化树分析比对为诺如病毒GⅡ.4亚型Sydney-2012变异株,片段长度为282 bp,核苷酸同源性为98%。结论引起大连市一起儿童急性肠胃炎疫情的诺如病毒是GⅡ.4亚型Sydney-2012变异株,此亚型也是目前引起儿童急性肠胃炎疫情的主要基因亚型。Objective To analyze the norovirus gene subtypes in children with acute gastroenteritis in Xigang district, Dalian city of China in October 2020. Methods The children′s fecal specimen were extracted for norovirus RNA and reverse transcription-polymerase chain reaction(RT-PCR) was used to amplify the norovirus subtypes.Lasergen 7.1, Norovirus Typing Tool Version 2.0, Clustal X 2.1, MEGA 10.1 and other software were used to construct neighbor-joining phylogenetic tree based on the gene sequence fragments of the virus strain and perform germline evolution analysis. Results Eleven specimens were positive for NV GⅡ nucleic acid, one of them was amplified by RT-PCR, the product was sequenced and compared with a Sydney-2012 variant of norovirus-based GⅡ.4 subtype, fragment length was 282 bp and the nucleotide homology was 98%. Conclusions The norovirus isolated from children with acute gastroenteritis in Xigang district,Dalian city in 2020 was subtype of GⅡ.4 Sydney-2012.
