摘  要:目的通过测定新生大鼠缺氧缺血性脑损伤(HIBD)脑组织中死亡蛋白激酶1(DAPK1)mRNA及其蛋白的表达,探讨死亡蛋白激酶1与缺氧缺血性脑损伤的关系.方法SD新生大鼠60只,随机分为模型组(HIBD组)和假手术组两组,其中HIBD组50只,根据处死时间不同分为5组(6 h、12 h、24 h、48 h和72 h各10只),假手术组10只,使用qRT-PCR、Western blot分别检测假手术组及HIBD组DAPK1 mRNA及蛋白的表达水平.结果假手术组DAPK1 mRNA和DAPK1蛋白均低表达,HIBD组均高表达,且HIBD不同时间点表达水平均较假手术组增高,差异有统计学意义(P<0.01);HIBD组48 h DAPK1 mRNA和DAPK1蛋白达到高峰.结论缺氧缺血性脑损伤可以使DAPK1的表达上调.Objective To determined the expression of death associated protein kinase 1(DAPK1) mRNA and its protein in the brain tissue of neonatal rats with hypoxic-ischemic brain damage(HIBD), and to explore the relationship between death associated protein kinase 1 and hypoxic-ischemic brain damage.Methods Sixty SD newborn rats were randomly divided into model group(HIBD group) and sham operation group, 50 rats in the model group were divided into 5 groups according to the time of execution(6 h, 12 h, 24 h, 48 h, 72 h), and 10 in sham operation group. The expression levels of DAPK1 mRNA and its protein were detected by qRT-PCR and Western blot.Results The expression level of DAPK1 mRNA and DAPK1 protein in sham operation group was lower than that in HIBD group(P<0.01), the expression of DAPK1 mRNA and its protein in HIBD group reached the peak at 48 hours after the injury.Conclusions The expression of DAPK1 was enhanced after hypoxic-ischemic brain injury.
