摘 要:从采自云南省嵩明县梁王山的高原线虫草(Ophiocordyceps highlandensis Zhu L. Yang&J. Qin)子实体中分离1株重寄生菌JZ025,根据形态特征观察和ITS序列分子生物学分析,确定其分类地位;设计3种不同固体培养基,筛选其最佳固体培养条件。结果表明,该重寄生菌为中国多头霉[Polycephalomyces sinensis (Q.T. Chen, S.R. Xiao&Z.Y. Shi) W.J. Wang, X.L. Wang, Y. Li, S.R. Xiao&Y.J. Yao]。在3种配方中,最适合菌株JZ025生长的固体培养条件为每瓶大米45 g,加58.5 mL液体培养基(葡萄糖5 g、蛋白胨15 g、KH2PO41.5 g、MgSO40.75g,加水至1 000 mL,pH自然)。The strain JZ025 was isolated from the spiece of Ophiocordyceps highlandensis which was collected in Liangwang Mountain of Yunnan province, then these one selected samples was identified by morphological characters and ITS sequence data analysis, and use three kinds of solid cultured medium to select the best culture conditions. The results showed that JZ025 was respectively identified as Polycephalomyces sinensis(Q.T. Chen, S.R. Xiao&Z.Y. Shi) W.J. Wang, X.L. Wang, Y. Li, S.R.Xiao&Y.J. Yao. Among the three kinds of solid culture medium, the suitable media for mycelia growth of JZ025 was 45 g rice of each bottle and 58.5 mL of liquid medium(glucose 5 g, peptone 15 g, KH2PO41.5 g, MgSO40.75 g, add water to 1 000 mL, pH natural).
