蛭龙活血通瘀胶囊对家兔颈动脉粥样硬化斑块组织p-NF-κB、NLRP3表达的影响

摘　　要：目的:探讨蛭龙活血通瘀胶囊对家兔颈动脉粥样硬化斑块组织p-NF-κB、NLRP3的影响。方法:将40只雌雄各半的清洁级新西兰大白兔随机分为正常组、模型组、蛭龙活血通瘀胶囊组、阿托伐他汀组、蛭龙活血通瘀胶囊+阿托伐他汀组,每组8只。各组经适应性普通饲料喂养1周后,除正常组外其余各组行右侧颈动脉内膜空气干燥术,术后继续高脂饲料喂养4周建立兔颈动脉粥样硬化模型,正常组普通饲料喂养。实验第5周末处死家兔,检查各组家兔血脂、颈动脉p-NF-κB、NLRP3蛋白表达,观察颈动脉病变情况。结果:与模型组比较,蛭龙活血通瘀胶囊和阿托伐他汀均可降低模型家兔血清总胆固醇(total cholesterol,TC)和甘油三酯(triglyceride,TG)(P<0.01),降低p-NF-κB、NLRP3蛋白在颈动脉组织中的表达(P<0.01),不同程度改善颈动脉病变情况,且蛭龙活血通瘀胶囊+阿托伐他汀组作用更明显。结论:蛭龙活血通瘀胶囊可降低p-NF-κB、NLRP3蛋白在高脂血症颈动脉粥样硬化家兔颈动脉组织中的表达,降低模型家兔血脂,改善颈动脉粥样硬化,且与阿托伐他汀联用效果更佳。Objective:To explore the effects of Zhilong Huoxue Tongyu capsules on the expressions of p-NF-κB and NLRP3 in carotid atherosclerotic plaque in rabbit.Methods:Forty New Zealand white rabbits,half male and female were randomized into the normal group,the model group,Zhilong Huoxue Tongyu capsules group,atorvastatin group,Zhilong Huoxue Tongyu capsules+atorvastatin group,eight rabbits each group.After one week of adaptive common diet,all the groups received air drying of the right carotid artery intima except the normal group after the operation,the rabbits were fed with high-fat diet for four weeks to establish carotid atherosclerosis model,the normal group were fed with common diet.The rabbits were sacrificed by the end of the 5th week,to detect the levels of blood lipid,the expressions of p-NF-κB and NLRP3 in carotid artery in different groups,to observe pathological changes of carotid artery.Results:Compared with the model group,Zhilong Huoxue Tongyu capsules and atorvastatin could lower the levels of TC and TG in rabbit models(P<0.01),lower the expressions of p-NF-κB and NLRP3 protein in carotid artery(P<0.01),improve carotid artery disease to different degrees,and the effects of Zhilong Huoxue Tongyu capsules+atorvastatin group the most notable.Conclusion:Zhilong Huoxue Tongyu capsules could lower the expressions of p-NF-κB and NLRP3 protein in carotid artery in atherosclerotic rabbits,decrease the levels of blood lipid in rabbit models,improve carotid atherosclerosis,and its effects the best in its combination with atorvastatin.
