化痰散结方对人胃癌MKN45细胞的体内外抑制作用

摘　　要：目的:观察化痰散结方对人胃癌MKN45细胞及其裸鼠移植瘤的生长抑制作用。方法:应用CCK-8法检测化痰散结方对体外胃癌MKN45细胞的生长抑制作用。建立裸鼠MKN45人胃癌移植瘤模型,并随机分为模型组、中药组和5-氟尿嘧啶(5-fluorouracil,5-fu)组,模型组灌胃生理盐水0.02 mL/g,中药组灌胃化痰散结方汤剂0.02 mL/g,5-fu组腹腔注射5-fu 25 mg/kg。4周后取出瘤块称移植瘤重量,观察化痰散结方对移植瘤的影响。结果:化痰散结方含药血清具有体外抑制人胃癌MKN45细胞增殖的作用,随药物作用浓度和时间的增加,抑制率也增加。化痰散结方可抑制裸鼠移植瘤的生长,中药组与5-fu组的抑瘤率分别为44.53%和57.81%。结论:化痰散结方可体外抑制MKN45细胞增殖,体内抑制MKN45细胞移植瘤生长。Objective:To observe the inhibitory effects of Huatan Sanjie prescription on human gastric cancer MKN45 cells and the growth of transplanted tumor in nude mice.Methods:CCK-8 method was used to detect the inhibitory effects of Huatan Sanjie prescription on human gastric cancer MKN45 cells in vitro.After establishing nude mice models with transplanted tumor of human gastric cancer MKN45 cells,the models were randomized into the model group,herbal group and 5-fu group,the model group were drenched with physiological saline,0.02 mL/g,herbal group with the decoction of Huatan Sanjie prescription,0.02 mL/g,and 5-fu group with 5-fu,25 mg/kg.After removing the tumor four weeks later,the weight of transplanted tumor was measured,to observe the effects of Huatan Sanjie prescription on transplanted tumor.Results:The serum containing Huatan Sanjie prescription could inhibit the proliferation of human gastric cancer MKN45 cells in vitro,the inhibitory rate increased with the increase of the concentrations and time.Huatan Sanjie prescription could inhibit the growth of transplanted tumor in nude mice,tumor-inhibition rates of herbal group and 5-fu group were 44.53% and 57.81% respectively.Conclusion:Huatan Sanjie prescription could inhibit the proliferation of MKN45 cells in vitro and the growth of MKN45 cells transplanted tumor in vivo.
