摘 要:从白色念珠菌的增殖、活性氧(ROS)的积累、细胞膜的完整性和线粒体功能障碍等几个方面,研究了香雪兰精油的抗真菌作用.结果表明:白色念珠菌对香雪兰精油极度敏感,精油的最小抑菌浓度和最小杀菌浓度分别为0.1%和0.25%(质量分数);精油处理能导致白色念珠菌核酸、蛋白质泄漏,细胞形态改变以及胞内ROS积累,进而加强脂质过氧化;氧化还原失衡是由抗氧化酶活性和线粒体膜电位的降低引起的.香雪兰精油对白色念珠菌具有良好的抑制作用,在抗真菌临床应用中具有很大的潜力.Infection of Candida albicans always caused serious effects on human health,while the long-term use of common anti-fungal medicine had side effects that limited their usage.In this paper,the anti-fungal effects of essential oil from Freesia hybrida were evaluated through proliferation of Candida albicans,ROS accumulation,integrity of cell membrane and dysfunction of mitochondria.The results showed that the minimum inhibitory concentration and minimal fungicidal concentration of essential oil were 0.1%and 0.25%,respectively.There was no obvious growth of Candida albicans at MIC concentration within 20 h.A large amount of ROS is accumulated,resulting in lipid peroxidation and nucleic acid and protein leakage.Further study showed that the redox imbalance was induced by the damage of antioxidant enzyme activity and the decrease of the mitochondrial membrane potential.These results suggest that the essential oil from Freesia hybrida had a good inhibitory effect on Candida albicans and had a great potential in antifungal clinical usage.
