摘 要:目的:观察黄精对联合诱导肝纤维化模型大鼠肝脏的保护作用及对TGF-β1、ICAM-1基因表达的影响。方法:将48只SPF级SD雄性大鼠随机分为4组:正常对照组、模型组及黄精中剂量组、黄精高剂量组,每组10只,正常对照组大鼠自由进食标准鼠粮、饮用清洁水源,其余各组大鼠采用皮下注射四氯化碳(CCl 4)、饮用20%酒精及喂养高脂低蛋白饮食方法联合诱导建立大鼠肝纤维化模型,第6周末观察病理切片确认造模成功后,各治疗组给予相应药物灌胃治疗6周,正常对照组和模型组给予等量生理盐水。HE染色观察肝脏结构病理变化;ELISA法检测血清中TGF-β1、ICAM-1水平;RT-PCR检测肝脏TGF-β1、ICAM-1基因表达水平的变化。结果:病理结果显示黄精中、高剂量组肝脏损伤与模型组相比明显减轻,肝细胞坏死和脂肪变性明显减少,纤维化程度减轻、炎细胞浸润情况有所改善;与模型组相比,血清中TGF-β1、ICAM-1含量和基因表达水平明显下降(P<0.01)。结论:黄精对联合诱导肝纤维化模型大鼠有保护作用,其机制可能与其对TGF-β1和ICAM-1水平的调节有关。Objective:To observe the protective effect of Polygonatum sibiricum on liver and its influence to the expressions of TGF-β1 and ICAM-1 in rats with liver fibrosis induced by combined induction.Methods:40 SPF SD male rats were randomly divided into the normal control group and the model group and the Polygonatum sibiricum groups of medium-dose and high-dose,with 10 rats in each group.Rats in the normal control group were administrated with standard food and clean water,the rats in the other groups were induced to establish hepatic fibrosis model by subcutaneous injection of carbon tetrachloride(CCl 4),combined with drinking 20%alcohol and feeding high-fat and low protein diet.At the end of the sixth week,the pathological sections were observed to confirm the success of the model.The treatment groups were given corresponding interventions for 6 weeks,and the normal control group and the model group were given the same amount of normal saline.HE staining was used to observe the pathological changes of liver structure;ELISA was used to detect the serum levels of TGF-β1 and ICAM-1;RT-PCR was used to detect the gene expressions of TGF-β1 and ICAM-1 in liver.Results:The pathological results showed that the liver injury,hepatocyte necrosis and lipid degeneration were significantly reduced,as well as fibrosis degree and inflammatory cell infiltration were significantly improved in Polygonatum sibiricum groups compared to those in the model group.The serum levels of TGF-β1,ICAM-1 and their gene expressions were significantly decreased in Polygonatum sibiricum groups compared to those in the model group(P<0.01).Conclusion:Polygonatum sibiricum has protective effect on liver fibrosis model rats,and its mechanism may be related to the regulation of TGF-β1 and ICAM-1 levels.
