摘 要:目的:采用网络药理学的方法探讨山药治疗糖尿病心肌病的作用机制。方法:在TCMSP数据库中寻找山药的活性成分,并对其进行靶点预测;在疾病数据库检索糖尿病心肌病(DCM)的靶点;利用Venn图得到山药和DCM的共同靶点;使用Cytoscape软件绘制“山药-活性成分-疾病-靶点”网络图;在STRING数据库中进行蛋白互作分析(PPI);利用DAVID数据库进行KEGG通路分析和GO生物功能注释。结果:发现了符合标准的16个活性物质和69个作用靶点,主要涉及ADRB2、ADRB1、NOS3等。在DAVID数据库分析中,获得了63个主要通路和130条GO生物功能注释。结论:山药可能通过多种成分、多个靶点、多条通路来治疗糖尿病心肌病,其作用机理可能是通过主要活性成分(-)-花旗松素和薯蓣皂苷元参与cGMP-PKG通路来调节氧化还原酶活性、凋亡过程的负调控等过程诱导高糖环境下的心肌细胞凋亡,从而防止心肌细胞肥大,进而治疗糖尿病心肌病。Objective:To explore the mechanism of Dioscoreae Rhizoma in treating diabetic cardiomyopathy based on network pharmacology.Methods:The active ingredients of Dioscoreae Rhizoma were found in TCMSP database and its target was predicted.The target of diabetic cardiomyopathy(DCM)was retrieved in disease database;the common target of Dioscoreae Rhizoma and DCM were obtained by the Venn diagram;network diagram of Dioscoreae Rhizoma-active ingredients-disease-target was drawn by using Cytoscape software.Protein-protein interaction(PPI)was analyzed in STRING database and KEGG pathway analysis and GO biological function were annotated by using DAVID database.Results:16 active substances and 69 targets were found,which mainly involved ADRB2,ADRB1,NOS3 and so on.In DAVID s database analysis,63 main pathways and 130 GO biological function annotations were obtained.Conclusion:Dioscoreae Rhizoma may be used to treat DCM through multiple ingredients,multiple targets and multiple pathways.The mechanism of action may be through the main active ingredients(-)-Taxifolin and Diosgenin participating in the cGMP-PKG pathway to regulate oxidoreductase activity,negative regulation of apoptosis process and other processes to induce cardiomyocyte apoptosis under high glucose environment,thereby preventing cardiomyocyte hypertrophy in the treatment of DCM.
