加味黑布膏对大鼠痤疮模型组织TLR2、TLR4表达的影响及其作用机制探讨

摘　　要：目的:观察加味黑布膏对大鼠耳廓复合痤疮模型组织Toll样受体(TLR)2、TLR4表达水平的影响,初步探讨其治疗轻度增生性痤疮瘢痕的作用机制。方法:抽取10只大鼠为空白组,剩余大鼠参照Kligman法建立大鼠耳廓复合痤疮模型后随机被分为加味黑布膏低、中、高剂量组,阳性对照组和模型组,治疗21d后,观察治疗前后皮损变化、大鼠耳廓造模区域组织病理变化,并以RT-PCR及Western Blot测定大鼠耳廓造模区域组织TLR2、TLR4的表达水平。结果:加味黑布膏高剂量组皮损已基本接近空白组,其余各治疗组皮损均有不同程度的改善,其中加味黑布膏高剂量组皮肤角质层接近空白组;RT-PCR及Western Blot检测:模型组耳廓组织TLR2、TLR4的表达显著高于空白组(P<0.01);除加味黑布膏低剂量组外,其他3组耳廓组织TLR2、TLR4的表达均低于模型组(P<0.05);加味黑布膏高剂量组优于阳性对照组(P<0.05)。结论:加味黑布膏对轻度增生性痤疮瘢痕有较好的治疗作用,其有效性可能与降低局部组织TLR2、TLR4的表达水平有关。Objective:To observe the effects of modified Heibu Ointment on the expression of TLR2 and TLR4 in rat auricular complex acne model,and to explore the mechanism of modified Heibu Ointment in the treatment of mild hyperplastic acne scar.Methods:Ten rats were selected as blank group.The remaining rats were randomly divided into low,medium and high-dose modified Heibu Ointment groups,positive control group and model group.After 21 days of treatment,the changes of skin lesions and histopathological changes of auricle model area were observed before and after treatment.RT-PCR and Western Blot were used to detect expression levels of TLR2 and TLR4.Results:The skin lesions in the high-dose group of modified Heibu Ointment were basically close to the blank group,and the other treatment groups had different degrees of improvement.Histopathological results:modified Heibu Ointment could improve the elongation and keratosis of epidermal process,reduce inflammatory cell infiltration and intercellular edema,and inhibit the growth of fibroblasts.The expression of TLR2 and TLR4 in auricle tissue of model group was significantly higher than that of blank group(P<0.01);the expression of TLR2 and TLR4 in auricle tissue of other three groups was lower than that of model group(P<0.05);the expression of TLR2 and TLR4 in auricle tissue of high dose group was better than that of positive control group(P<0.05).Conclusion:Modified Heibu Ointment has a better therapeutic effect on mild hypertrophic acne scar.Its effectiveness may be related to the decrease of TLR2 and TLR4 expression in local tissues.
