摘 要:目的:基于复方雷公藤汤最佳配比剂量正交设计考察其对肾病大鼠肝脏的减毒增效作用。方法:采用一次性注射阿霉素至大鼠尾静脉,复制微小病变肾病综合征模型。采用正交实验法进行设计,将肾病模型大鼠随机分为9组,每组10只,连续灌服不同剂量配比的复方雷公藤汤14d。增效采用24h尿蛋白定量检测,对肝脏减毒测量谷丙转氨酶、谷草转氨酶。筛选出复方雷公藤汤最佳剂量配比组合。结果:(1)正交设计对肾病大鼠肝脏的减毒实验结果:影响因素大小排序为雷公藤>甘草>制何首乌>黄芪,其中8号复方效果最好,最优配比为A_(3)B_(2)C_(1)D_(3)。(2)经正交设计对降低肾病大鼠24h尿蛋白实验结果:影响因素大小排序为雷公藤>甘草>黄芪,制何首乌无显著影响,其中3号复方效果最好,最优配比为A_(1)B_(3)C_(3)D_(3)。结论:复方雷公藤汤治疗肾病时如以减低肝脏毒性为主,采用A_(3)B_(2)C_(1)D_(3)配比组成复方,治疗肾病时如以降低24h尿蛋白为主,采用A_(1)B_(3)C_(3)D_(3)配比组成复方,两种配比复方均起到减毒增效作用。Objective:Based on the orthogonal design of the best ratio and dosage of Compound Tripterygium Wilfordii Decoction,the attenuation and synergism of Compound Tripterygium Wilfordii Decoction on the liver of rats with nephropathy were investigated.Methods:The model of minimal change nephrotic syndrome was established by injecting adriamycin into the tail vein of rats once.The kidney disease model rats were randomly divided into 9 groups,10 rats in each group,and were given Compound Tripterygium Wilfordii Decoction with different doses for 14 days.24-hour urine protein quantitative detection was used for synergism,and glutamic pyruvic transaminase and glutamic oxaloacetic transaminase were measured for liver attenuation.The best dosage combination of Compound Tripterygium Wilfordii Decoction was screened out.Results:(1)Experimental results of liver attenuation by orthogonal design in nephrotic rats:the order of influencing factors is Tripterygium wilfordii>Glycyrrhiza uralensis Fisch>Radix Polygoni Multiflori Preparata>Astragalus membranaceus,among which compound No.8 has the best effect,and the optimal ratio is A_(3)B_(2)C_(1)D_(3).(2)The experimental results of reducing 24-hour urinary protein in nephropathy rats by orthogonal design:the order of influencing factors is tripterygium wilfordii>licorice>astragalus membranaceus,and the prepared Polygonum multiflorum has no significant effect,among which compound No.3 has the best effect,and the optimal ratio is A_(1)B_(3)C_(3)D_(3).Conclusion:Compound Tripterygium Wilfordii Decoction is mainly used to reduce hepatotoxicity when treating nephropathy,and A_(3)B_(2)C_(1)D_(3) is used to make compound,while A_(1)B_(3)C_(3)D_(3) is used to make compound,which has the effects of reducing toxicity and increasing efficiency.
