摘  要:目的:观察复方苦参汤对溃疡性结肠炎(UC)小鼠模型结肠黏膜诱导型一氧化氮合酶(iNOS)和环氧化酶2(COX-2)蛋白的作用机制。方法:将20只BALB/c小鼠随机分为4组:正常组、模型组、复方苦参汤组和美沙拉嗪组,每组5只。先适应性喂养1周,在第8天,除正常组外,其余3组分别予3%葡聚糖硫酸钠(DSS)自由饮用1周制备实验性UC小鼠模型。同时在造模第1天开始,正常组和模型组每日均以蒸馏水灌胃,复方苦参汤组及美沙拉嗪组每日分别予复方苦参汤、美沙拉嗪灌胃,连续治疗1周。取各组结肠组织固定后HE染色病理观察,采用Western Blot检测各组小鼠结肠组织iNOS及COX-2蛋白的表达。结果:与正常组比较,模型组iNOS及COX-2蛋白表达量显著升高(P<0.01);与模型组比较,两给药组iNOS及COX-2蛋白表达量显著降低(P<0.01)。结论:复方苦参汤治疗UC的作用机制可能与抑制结肠组织i NOS及COX-2蛋白表达有关。Objective:To observe the mechanism of Compound Kushen Decoction on inducible nitric oxidesynthase(i NOS)and cyclooxygenase 2(COX-2)proteins in the colonic mucosa of ulcerative colitis(UC)mice.Methods:Twenty BALB/c mice were randomly divided into 4 groups:normal group,model group,Compound Kushen Decoction group and mesalazine group,with 5 mice in each group.After one week of adaptive feeding,on the 8 th day,except for the normal group,the other three groups were given 3%dextran sulfate sodium(DSS)for one week to prepare a mouse model of experimental ulcerative colitis.At the same time,on the first day of modeling,the normal group and the model group were given daily gavage with distilled water,the Compound Kushen Decoction group and the mesalazine group were given daily gavage with Compound kushen Decoction and mesalazine,respectively for one week.The colonic tissues of each group were taken for pathological observation after fixation with HE staining examination.Western Blot was used to detect the expression of iNOS and COX-2 protein in colon tissue of each group.Results:Compared with the normal group,the expression levels of iNOS and COX-2 protein in the model group were significantly increased(P<0.01).Compared with the model group,the expression of iNOS and COX-2 protein in the Compound Kushen Decoction group and mesalazine group were significantly decreased(P<0.01).Conclusion:The mechanism of Compound Kushen Decoction in the treatment of ulcerative colitis may be related to the inhibition of iNOS and COX-2 protein expression in colon tissue.
