摘 要:目的:探讨防风对桃红四物汤活血化瘀治疗肝纤维化的增效作用及其机制。方法:清洁级雄性SD大鼠随机分为正常对照(A)组8只,造模组42只。CCl4大豆油溶液诱导肝纤维化模型。将造模组随机分为模型组、秋水仙碱组、防风桃红四物汤组,桃红四物汤组及防风组。检测丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、碱性磷酸酶(ALP)、透明质酸(HA)、层黏连蛋白(LN)、Ⅲ型前胶原蛋白(PCⅢ)、Ⅳ型胶原蛋白(Ⅳ-C)的变化。Western Blot及免疫组织化学法检测转化生长因子-β1(TGF-β1)、α平滑肌肌动蛋白(α-SMA)的表达。结果:与秋水仙碱组比较,防风桃红四物汤组大鼠血清中的ALT、AST、HA、PCⅢ、Ⅳ-C及肝组织中TGF-β1及α-SMA蛋白的表达量均显著下调(P<0.05)。结论:防风具有增效桃红四物汤活血化瘀的作用,其机制可能与下调TGF-β1及α-SMA蛋白的表达有关。Objective:To explore the synergistic effect and mechanism of Radix Saposhnikoviae on hepatic fibrosis treated by Taohong Siwu Decoction.Methods:SD rats were randomly divided into normal control group(n=8)and model group(n=42).Hepatic fibrosis models were induced by CCl4 soybean oil solution and randomly divided into pathological model group,colchicine group,Radix Saposhnikoviae Taohong Siwu Decoction group,Taohong Siwu Decoction group and Radix Saposhnikoviae Decoction group.The changes of AST,ALT,ALP,HA,LN,PCⅢand IV-C were detected.The expression of TGF-β1 andα-SMA was detected by Wstern Blot and immunohistochemistry.Results:Compared with group C,ALT,AST,HA,PCⅢ,IV-C in serum and TGF-β1 andα-SMA protein expression in liver tissues of group D rats were significantly decreased(P<0.05).Conclusion:Radix Saposhnikoviae can promot blood circulation and remove blood stasis of Taohong Siwu Decoction,and its mechanism may be related to down-regulating TGF-β1 andα-SMA.
