摘  要:目的:考察木犀草素对胶原诱导型关节炎(CIA)大鼠NLRP3炎性小体调节作用,观察木犀草素对动物模型关节骨保护表现。方法:随机选取雌性SD大鼠分为空白组、模型组、木犀草素100mg/kg组(木犀草素组)、木犀草素50mg/kg+MCC95020mg/kg组(联合给药组)。除空白组外,其余各组构建关节炎模型。连续给药5周并测量大鼠足跖容积,免疫组化法检测骨组织内骨保护素(OPG)表达,免疫蛋白印迹法检测足跖内NLRP3、胱天蛋白酶-1(Caspase-1)、RANKL、血管内皮生长因子(VEGF)、低氧诱导因子-1α(HIF-1α)蛋白表达,ELISA试剂盒检测血浆白介素(IL)-1β和IL-18表达。结果:与模型组比较,木犀草素组、联合给药组大鼠足跖肿胀度显著降低(P<0.01);免疫组化显示木犀草素组、联合给药组骨组织内OPG蛋白表达较高(P<0.01);免疫蛋白印迹显示木犀草素组、联合给药组关节滑膜内NLRP3、Caspase-1、RANKL、VEGF和HIF-1α蛋白表达下调(P<0.01,P<0.05);ELISA显示木犀草素组、联合给药组血浆IL-1β、IL-18表达下调(P<0.01)。结论:木犀草素可抑制类风湿关节炎NLRP3炎性小体的表达,进一步降低足跖内Caspase-1、RANKL、VEGF和HIF-1α蛋白表达并增强骨组织内骨保护素OPG蛋白表达,从而起到骨关节保护作用。Objective:To investigate the regulatory effect of luteolin on NLRP3 inflammatory corpuscle in collagen induced rheumatoid arthritis(CIA)rats,and to observe the protective effect of luteolin on joint bone in rheumatoid arthritis rats.Methods:Female SD rats were randomly divided into control group,model group,luteolin 100 mg/kg group(luteolin group)and luteolin 50 mg/kg+MCC95020 mg/kg group(combined administration group).The model was established in all groups except control group.After five weeks of administration,the voix pedis volume in rats was measured.The OPG protein expression in bone tissue was detected by immunohistochemical method.Western Blot study NLRP3,Caspase-1,RANKL,VEGF and HIF-1αprotein in voix pedis,and IL-1βand IL-18 expression in plasma were detected by ELISA kit.Results:Compared with the model group,the swelling degree of voix pedis in luteolin group and combined administration group were decreased significantly(P<0.01).The immunohistochemistry showed that the expression of OPG protein in bone tissue was up-regulated in luteolin group and combined administration group.The Western Blot showed that the expression of NLRP3,Caspase-1,RANKL,VEGF and HIF-1αprotein were down-regulated in luteolin group and combined administration group(P<0.01,P<0.05).ELISA showed that the expression of plasma IL-1βand IL-18 were down-regulated in luteolin group and combined administration group(P<0.01).Conclusion:Luteolin may inhibit the the expression of NLRP3 inflammatory corpuscle of rheumatoid arthritis rats,further decrease the expression of Caspase-1,RANKL,VEGF,HIF-1αprotein in voix pedis and enhance the expression of OPG protein in bone tissue,so as to play a role in bone joint protection.
