摘  要:为保护和利用花生种质资源,对花生RAPD反应条件进行了优化及遗传多样性分析。优化后的RAPD总反应体积为20μL,其中,2×Taq PCR Mix 10μL、引物1.6 pmol、模板20 ng。利用优化后的反应条件和筛选出的24个引物在22份花生样品中扩增得到了134条清晰的电泳条带,其中,多态性条带数为112条,多态性百分率为83.58%,平均每个引物产生多态性条带数为4.667条。通过分子标记RAPD进行遗传多样性分析,将现在北方地区种植的22份花生种质资源划分为3大类群,对北方地区花生生产的种植结构以及新品种选育具有重要的参考价值。To protect and utilize the germplasm resources of Arachis hypogaea,RAPD reaction system of A.hypogaea was optimized and its genetic diversity was analyzed.The results showed that total volume of optimal RAPD reaction was 20μL,including 10μL of 2×Taq PCR Mix,1.6 pmol of primer,20 ng of template DNA.Using the optimal RAPD reaction,134 clear electrophoresis bands were amplified from 22 samples of A.hypogaea with 24 primers,in which there were 112 polymorphic bands,the percentage of polymorphic percentage was 83.58%and each primer amplified an average number of 4.667 polymorphic bands.The 22 germplasm resources of A.hypogaea planted in northern China was divided into 3 groups,which provide a valuable base for planting structure and breeding of new varieties of A.hypogaea in northern China.
