摘  要:采集乌鲁木齐某马术俱乐部226匹纯血马鼻拭子样品,用高通量测序检测纯血马携带病毒多样性,结果宏病毒组学分析共产生73664946条reads,有16067条reads(0.022%)注释为哺乳动物病毒,其中105条reads注释到马疱疹病毒5型(EHV-5)gH基因。根据GenBank中的EHV-5参考株2-141/67 gH基因序列,设计特异性引物,验证宏病毒组学结果,并对gH基因的氨基酸序列进行遗传进化分析。结果14匹健康纯血马鼻拭子样品呈EHV-5阳性,阳性率为6.2%(14/226),序列分析表明,鉴定的14株EHV-5 gH基因的核苷酸和氨基酸序列与澳大利亚、意大利及日本分离株gH基因的核苷酸和氨基酸序列同源性分别为98.4%~100%和90.3%~100%。遗传进化分析显示我国14株EHV-5与澳大利亚2-141/67和EHV-5.2-141株亲缘关系较近,而与意大利glycoprotein H株、日本16-I-1064和1-I-790株处于不同分支。This study collected 226 equine nasal swab samples from Wulumuqi City of Xinjiang,and used high-throughput sequencing to detect viral diversity carried by horses.Virome analysis revealed that nasal swab samples generated 73664946 reads,with 16067 of them(0.022%)annotated to mammalian viruses,and 105 reads were annotated to equine herpesvirus 5(EHV-5).According to EHV-52-141/67 reference strain,specific primers were designed to verify the results of viral metagenomics.PCR detection results showed that EHV-5 gH gene was positive in the nasal swab samples of 14 healthy thoroughbred horses,the positive rate was 6.2%(14/226).Sequence analysis showed that the homology of nucleotides and amino acids among our 14 EHV5 gH was 98.2%-100%and 95.3%-100%,respectively.The homology of gH nucleotides and amino acids identified in our study was 98.4%-100%and 90.3%-100%,respectively,with Australian,Italian and Japanese isolates.Phylogenetic analysis on the basis of gH amino acids showed that Chinese EHV5 gH was in the same branch with Australian 2-141/67 and EHV-5.2-141 strains,and but in different branches with Italian glycoprotein H strain,Japanese 16-I-1064 and 1-I-790 strains.In the current study,EHV-5 was first reported in horse population of China.This study identified EHV-5 and took genetic evolution analysis on its gH gene.
