摘 要:选用甘肃省道地药材红芪作为试验材料,通过水提醇沉法提取红芪多糖,并以单因素试验与响应面法相结合的方法优化红芪多糖的提取工艺。单因素试验设置提取温度、提取时间、提取次数、液料比和醇沉比5个因素,每个因素设置5个水平。在此基础上以多糖提取率为衡量指标,进行Box-Behnken试验设计,通过回归模型的建立及显著性检验,得到红芪多糖提取工艺的最佳参数,再利用该工艺参数条件进行验证。结果显示,红芪多糖的最佳提取条件为提取温度为80℃,提取时间为3 h,提取次数为4次,液料比为17 mL/g,醇沉比75%。最佳条件下红芪多糖的平均提取率为7.70%,与理论提取率8.01%无显著性差异(P>0.05)。证明所建立的数学模型是合理的,认为采用单因素试验结合响应面法优化红芪多糖的工艺稳定可行,为红芪多糖的有效提取提供理论依据,为进一步分离纯化红芪多糖奠定基础。In this experiment,the genuine Radix Hedysari in Gansu province as experimental material was used to extract RHPS by water-extraction and alcohol-precipitation method.The extraction process was optimized by a combination of single factor experiment and response surface method.The single factor test was set in five factors,including extraction temperature,extraction time,extraction times,liquid-to-material ratio and alcohol precipitate ratio,and each factor was set in five levels.Based on this,the Box-Behnken experiment design was performed with the polysaccharide extraction yield as the index.The optimal parameters of the extraction process of RHPS were obtained through the establishment of regression model and significance test.And then the verified experiment was performed.The results showed that the optimal extraction conditions were as following:extraction temperature of 80℃,extraction time of 3 h,extraction times of 4 times,liquid-to-material ratio of 17 mL/g,and alcohol precipitate ratio of 75%,respectively.Under the optimal extraction conditions,the average extraction yield was 7.70%,which was not significantly different from the theoretical extraction yield of 8.01%(P>0.05).It is demonstrated that the established models were proved to be reasonable.The combination of single factor experiment and response surface method used to extract RHPS was stable and feasible.It provided a theoretical basis for the effective extraction of RHPS,and laid a foundation for the further separation and purification.
