摘 要:以草珊瑚种子作为外植体,研究了赤霉素和NaOH浸种、升汞处理时间长短、不同培养基和植物生长调节剂对草珊瑚种子离体培养的影响。结果表明:0.02%赤霉素浸种24 h不仅可以提高草珊瑚种子萌芽率,还可以缩短萌发时间;采用0.1%升汞处理种子3 min,种子的污染率仅1.17%;草珊瑚种子初代培养的最佳培养基为1/4 MS+0.5 mg/L 6-BA+0.05 mg/L NAA+30 g/L蔗糖+5 g/L琼脂,萌芽率达78.87%;最佳增殖培养基为MS+4.0 mg/L 6-BA+0.5 mg/L NAA+30 g/L蔗糖+5 g/L琼脂,丛生芽诱导率达82.96%。Seeds were used as explants to study the effects of the immersion of GA3 and NaOH,the duration of mercury-rising treatment,different media and plant growth regulator on the in vitro culture of Sarcandra glabra.The results showed that immersion 24 hours with 0.02%GA3 could not only improve seed germination rate,but also shorten germination time;The contamination rate of seeds was only 1.17%with 0.1%HgCl2 treatment for 3min;The best culture medium for the primary culture of seed was 1/4 MS+0.5 mg/L 6-BA+0.05 mg/L NAA+30g/L sucrose+5g/L agarophyte,the germination rate was 78.87%;The best medium for proliferation was MS+4.0mg/L 6-BA+0.5mg/L NAA+30g/L sucrose+5g/L agarophyte,and The induced rate of fascicled shoots reached 82.96%.
