不同部位镇坪黄连小檗碱、总生物碱提取与含量测定

摘　　要：为了研究镇坪黄连各部位药用价值,提升黄连资源利用效益。采用超声辅助回流和盐析法对不同部位黄连材料中总生物碱、小檗碱等药效成分进行提取和分离纯化。通过薄层色谱、紫外分光光度法和HPLC定性、定量分析测定了镇坪黄连不同部位样品中总生物碱和小檗碱含量。研究结果表明:不同部位镇坪黄连总生物碱和小檗碱含量高低次序为主根>须根>叶茎,其总生物碱含量为13.05%±0.94、3.05%±0.41和1.17%±0.04;小檗碱含量依次为5.70%-5.94%、1.71%-1.73%和1.06%-1.16%。由此可知镇坪黄连的主根、须根和叶茎中均含有大量的黄连生物碱和一定量的小檗碱,在目前主要以黄连主根为中药材原料利用的同时,应加强黄连须根、叶茎的利用力度,以提高优质黄连资源的经济效益。In order to study the medicinal value of various parts of Zhenping Coptis chinensis and enhance the utilization benefit of the Coptis chinensis resources.The total alkaloids and berberine in different parts of Coptis chinensis were extracted and purified by ultrasonic assisted reflux and salt precipitation method.The contents of total alkaloids and berberine in different parts of Coptis chinensis were qualitativly and quantitatively determined by TLC,UV spectrophotometry and HPLC.The results showed that the contents of total alkaloids and berberine in different parts of Coptis chinensis were main roots>fibrous roots>leaf stems,with total alkaloids content of 13.05%±0.94,3.05%±0.41 and 1.17%±0.04,and berberine content of 5.70%~5.94%,1.71%~1.73%and 1.06%~1.16%.It can be seen that the main root,fibrous root and leaf stems of Zhenping Coptis chinensis contain a large number of alkaloids and a certain amount of berberine.With the main root as the raw material of Chinese herbal medicine,the development and utilization fibrous roots and leaf stems should be strengthened to improve the economic benefit of Zhenping Coptis chinensis resources.
