摘 要:极长链脂肪酸延伸酶蛋白家族(elongation of very-long-chain fatty acids,ELOVLs)是合成脂肪酸过程中的重要限速酶,主要调控血糖、血脂及一些代谢疾病的发生。为探究宁夏地方品种静原鸡ELOVL5基因编码蛋白的结构与功能,本试验根据GenBank中已公布的原鸡ELOVL5基因序列,针对8个外显子区域设计特异性引物,经DNA混合池扩增、测序后,对其编码区进行功能生物信息学分析。结果表明,静原鸡ELOVL5基因编码区全长1023 bp,编码340个氨基酸;ELOVL5蛋白存在1个低复杂度保守序列及7个跨膜结构域,25个磷酸化位点,9个B细胞抗原表位,与acidPPc、BC10、RasGEFN蛋白存在部分重叠区域(41~142 aa,72~125 aa,207~336 aa),为非分泌型的稳定水溶性蛋白,空间结构以α-螺旋和无规则卷曲为主;亚细胞定位结果显示ELOVL5蛋白可能在线粒体内行使重要的生物学功能。该研究结果为后续ELOVL5蛋白功能的深入研究提供科学依据。The family of extremely long-chain fatty acid extension enzymes(elong-chain fatty acids)is an important speed limit enzyme in the process of synthesizing fatty acids,which regulates the occurrence of blood sugar,lipids and some metabolic diseases.In order to explore the structure and function of the gene coding protein of Jingyuan chicken ELOVL5 of Ningxia local species,this experiment designed specific foreign primers for 8 exon regions according to the gene sequence of the original chicken ELOVL5 published in GenBank,and carried out functional bioinformatics analysis of the coding area after amplification and sequencing of the DNA mixing pool.The results showed that the full-length was 1023 bp of ELOVL5 CDS sequence,and encoding 340 amino acids;ELOVL5 protein has 1 low complexity conservative sequence and 7 transmembrane domains,25 phosphorylation sites,9 B-cell antigen epitopes,partial overlap with acidPPc,BC10,RasGEFN protein(41 to 142 aa,72 to 125 aa,207 to 336 aa),a non-secretory stable water-soluble protein,the spatial structure dominated by Alpha-helix and random curl;Subcellular positioning results show that the ELOVL5 protein may perform important biological functions in the mitochondria.The results provide a scientific basis for the further in-depth study of ELOVL5 protein function.
