摘 要:植物内生菌与其宿主在长期进化过程中形成一种共生或互生的关系,对植物的生长发育、病虫害防治、植物抗性、生物修复等方面都具有积极的作用。为了解植物不同生态位内生菌菌群组成和形成机制,本研究以番茄为材料,通过高通量测序解析番茄植株根、茎和叶3个不同生态位内生菌菌群组成与结构差异。收集番茄根、茎、叶组织样品分别消毒,并分别进行研磨破碎提取基因组DNA,检验合格后送交公司进行16S rDNA序列V5~V7可变区的Illumina Miseq测序和微生物多样性分析。结果表明,植物植株内不同生态位内生细菌菌群结构组成和多样性有一定的差异。其中,根内生菌优势菌群主要由Proteobacteria、Actinobacteria、Firmicutes等组成;茎的优势菌群包括Proteobacteria和Firmicutes等;而叶的优势菌群主要由Proteobacteria等组成。Proteobacteria是番茄根、茎、叶中共有的主要优势内生菌群,但其在3个不同生态位中所占比例大小明显不同。根内生菌具有更高的种群多样性,且与茎和叶相比具有明显的差异性。总体来讲,根和茎内生菌组成和多样性相似度更高,Venn图分析表明根和茎有更多的共有OTU,而根和叶都有较多的特有OTU,表明茎的菌群受根的影响较大,而根和叶的菌群组成有较高的独立性。Plant endophyte,which plays a positive role in plant growth and development,pest control,plant resistance,bioremediation and other aspects,has formed a symbiotic or symbiotic relationship with their host in the long-term evolution process,In this paper,to understand movement of endophyts in plant tissues and their assembly mechanism of endophytic flora,the composition and structure of bacterial endophytic flora of tomato in different parts,roots,stems and leaves were analyzed by using the high-throughput sequencing method.The samples for tomato root,stem and leaf were separately collected and disinfected.The processed samples were ground and broken to extract their genomic DNA.The extracted genomic DNA samples were sent to the company for amplifying the V5-V7 region of bacterial 16 S rDNA and Illumina Miseq sequencing.The results showed that the plant tissues had certain effects on community structure and bacterial diversity of plant endophytic microbiome.Among them,Proteobacteria was the predominant micro flora of endophytes of tomato,and it is the main dominant micro flora shared by roots,stems and leaves.However,the proportions of Proteobacteria were obviously different in three different ecological niches.In addition,Actinobacteria and Firmicutes in tomato root,and Firmicutes in stem,were also among the dominant microflora.Root endophytes had higher population diversity and were obviously different from stem and leaf microbiome.In general,the composition and diversity of endophytic bacteria of roots and stems were more similar.Venn diagram analysis showed that roots and stems share more OTU,while roots and leaves have more unique OUT,suggesting the stem flora was greatly influenced by root,while the composition of root and leaf flora was relatively independent.
