摘 要:为了研究聚磷菌的分离、鉴定以及除磷特性,本研究通过富集培养、分离培养、MOPS(10×)培养、异染颗粒染色实验,并对CASS反应器中的活性污泥进行了聚磷菌的初筛和除磷能力复筛,得到一株除磷能力较强的菌株P2。结合形态学、生理生化试验和分子生物学鉴定试验对P2进行菌种鉴定以及行相关性能试验研究。结果表明:分离所得的菌株P2是一株柠檬酸杆菌,其除磷率可达到85%;菌株P2的生长迟缓期为0~6 h,对数生长期为6~24 h;菌株P2高效除磷的最适pH值为7,温度30℃,时间12 h,微量元素浓度为2 mL/L。菌株P2稳定期持续时间长,适应环境能力较强,对于各种环境下废水中磷的去除有较好效果。In order to study the separation,identification and phosphorus removal characteristics of phosphorus-accumulating,a bacterial strain P2 with strong phosphorus removal ability was obtained by accumulation culture,isolating culture,MOPS(10×)culture,metachromatic granular dyeing experiment,and primary screening of phosphorus-accumulating and re-screening of phosphorus removal ability from activated sludge in CASS reactor.The bacteria type of P2 was identified and the related properties of P2 were tested by morphology,physiological and biochemistry test and molecular biology identity test.The results showed that the bacterial strain P2 obtained by separation was a Citrobacter with a phosphorus removal rate of 85%.The growth lag phase of bacterial strain P2 was 0~6 h,and the logarithmic growth phase was 6~24 h.The optimum pH value for efficient phosphorus removal of bacterial strain P2 was 7,the temperature was 30℃,the time was 12 h,and the concentration of trace elements was 2 mL/L.Strain P2 has a long stable period and strong adaptability to the environment.It has a good effect on phosphorus removal from wastewater in various environments.
