摘  要:马铃薯(Solanum tuberosum)作为全球第三大粮食作物,在国内外广泛种植,但其生长经常受到各种逆境胁迫,使其正常生长发育和代谢受阻,对产量和品质的形成产生不利影响。抗坏血酸过氧化物酶(APX)作为一种清除活性氧的关键酶,在保护植物免受逆境胁迫发挥着重要作用。为了深入了解StAPX基因在马铃薯应激反应中的作用,本研究通过生物信息学方法从马铃薯基因组数据库中检索出了8个StAPXs基因,并对其染色体定位、二级结构、进化关系、保守功能域、启动子顺式作用元件和表达模式进行了分析。结果表明,马铃薯StAPXs基因编码的氨基酸数在250~420之间,分子量介于27.35~45.83 kD范围内,均为亲水性蛋白。亚细胞定位于过氧化酶体、细胞质、叶绿体和线粒体中。在染色体上呈现不均等分布。蛋白二级结构均主要由α螺旋和无规卷曲组成。系统进化分析发现,马铃薯StAPXs与番茄SlAPXs亲缘关系最近,其次是拟南芥,与水稻OsAPXs同源性较低。StAPXs基因外显子数目在8~12之间,氮端和碳端均含有UTR,编码的蛋白序列均含有Motif3和Motif5等保守基序。启动子顺势作用元件分析的结果显示,StAPXs中含有多个参与激素响应、干旱、低温、机械损伤及其它应激反应相关的顺式作用元件和转录因子结合位点。转录组数据分析表明,StAPXs基因在不同的组织中均有表达,特别是StAPX1、StAPX4、StAPX5、StAPX7的表达水平较其它基因高。另外,部分成员受盐、甘露醇、35℃高温、激素和生物胁迫等逆境的诱导表达。本研究结果提供了StAPXs基因家族成员的序列和部分功能信息,为进一步研究StAPXs基因参与逆境胁迫的分子机制提供参考。As the world’s third largest food crop,potato(Solanum tuberosum)is widely cultivated around the world.However,its growth is often subject to various stresses,which adversely affects the production and quality of potato.Aseorbate peroxidase(APX)plays an important role in protecting plants from stress.In order to further understand the role of,StAPXs genes in potato stress response,8 StAPXs genes were identified from potato genome database by bioinformatics method,and their chromosome location,secondary structure,evolutionary relationship,conservative functional domains,promoter cis-acting element and expression patterns were analyzed.The results showed that the number of amino acids encoded by StAPXs genes in potato ranged from 250 to 420 aa,and the molecular weight ranged from 27.35 to 45.83 kD,all of which were hydrophilic proteins.The subcellular localization of these proteins were found in peroxisome,cytoplasm,chloroplasts and mitochondria.The,StAPXs genes were unevenly distributed across potato chromosomes.The secondary structure of these proteins is mainly composed of alpha helix and random curl.Phylogenetic analysis showed that potato,StAPXs had the closest relationship with tomato SlAPXs,followed by Arahidopsis thaliana,and had lower homology with rice OsAPXs.The number of exons of StAPXs genes ranges from 8 to 12.Both the N and C terminals of these proteins contained UTR,and the encoded protein sequences contained Motif3,Motif5 and other conserved motifs.Analysis of the promoter homeopathic elements showed that StAPXs contained multiple cis-acting elements and transcription factor binding sites involved in hormone response,drought,low temperature,mechanical injury and other stress responses.Transcriptome data analysis showed that StAPXs genes were expressed in different tissues,and the expression levels of StAPX1,StAPX4,StAPX5 and StAPX7 were higher than other potato A PX genes.In addition,some members of StAPXs were induced by salt,mannitol,high temperature,hormones and biological stress.The results of
