摘  要:【目的】研究地黄烯醇化酶基因(RgENO)的生物信息学功能及其表达情况。【方法】利用高通量测序对混合地黄发育块根进行转录组测序,通过基因注释得到地黄烯醇化酶基因(RgENO)序列,RgENO为780 bp,编码260个氨基酸的蛋白质。利用生物信息分析软件对RgENO进行同源性比对、结构、跨膜结构区和信号肽分析;用RT-qPCR检测其空间表达。【结果】RgENO蛋白质相对分子量为27.7 Kda,理论等电点pI为7.69,具有烯醇化酶典型的C端与N端双结构域,属于TIM磷酸结合超家族,不具有跨膜结构和信号肽,与芝麻的烯醇化酶相似度约为98%;RgENO在地黄的根、茎和叶中均有表达,但差异不明显。【结论】RgENO的编码蛋白质与芝麻的烯醇化酶相似度最高,约为98%;其在地黄根、茎和叶中均有表达。【Objective】The paper is to study the bioinformatics function and expression situation of Rehmannia glutinosa enolase gene(RgENO).【Method】High-throughput sequencing is used to transcriptome sequence of mixed R.glutinosa developmental tubers,and then RgENO sequence with 780 bp and encoding a protein of 260 amino acids is obtained through gene annotation.Bioinformatics analysis software is employed to analyze the homology alignment,structure,transmembrane domain and signal peptide of RgENO.The spatial expression is detected by RT-qPCR.【Conclusion】The relative molecular weight of RgENO protein is 27.7 Kda with pI of 7.69.It has double domains with typical C-terminal and N-terminal of enolase,belongs to TIM phosphate binding superfamily,without transmembrane structure or signal peptide.The similarity to sesame enolase is about 98%.RgENO is expressed in root,stem and leaves of R.glutinosa with no obvious difference.【Conclusion】The similarity of RgENO encoded protein and sesame enolase is the highest of 98%.It is expressed in root,stem and leaves of R.glutinosa.
