摘 要:目的:研究川射干鸢尾苷元磺酸钠体外抗柯萨奇病毒和副流感病毒的效果,为抗病毒新药研发提供依据。方法:建立柯萨奇病毒感染Vero细胞、副流感病毒感染LLC-MK2细胞模型,以细胞增殖检测(CCK-8)法或细胞病变效应(CPE)法作为检测方法,了解药物对细胞的毒性以及体外抗病毒的效果,实验中以利巴韦林作为对照药物。结果:川射干鸢尾苷元磺酸钠对Vero细胞和LLC-MK2细胞的TC50浓度分别为2157.50μg/ml和1577.29μg/ml,川射干鸢尾苷元磺酸钠能明显抑制柯萨奇病毒和副流感病毒所致的细胞病变,IC50分别为114.05μg/ml和312.50μg/ml,治疗指数(TI)分别为18.92和5.05。结论:在体外抗病毒实验中,川射干鸢尾苷元磺酸钠显示出抑制柯萨奇病毒和副流感病毒的作用。Objective:Observing the effect of Tectorigenin sodium sulfonite against coxsackievirus and human parainfluenza virus in vitro to provide the basis of developing new antiviral drugs.Methods:Establishing models of coxsackievirus infecting vero cell and human parainfluenza virus infecting LLC-MK2cell.Using the method of CCK-8or CPE as the observation and detection method to study the drug toxicity to the cells and the antiviral effect.Using ribavirin as control medicine.Results:The TC50 of Tectorigenin sodium sulfonite to Vero cell and LLC-MK2cell were 2157.50μg/ml and 1577.29μg/ml respectively.Tectorigenin sodium sulfonite could inhibit the cell lesion induced by CVB2virus and HPIV2virus obviously,the IC50 were 114.05μg/ml and 312.50μg/ml respectively and the TI were 18.92and 5.05respectively.Conclusion:Tectorigenin sodium sulfonite shows the ability of anti-coxsackievirus and anti-HPIV in vitro antiviral experiment.
