摘 要:为探讨白介素2(interleukin-2,IL-2)在中国黄羽鹌鹑脾脏中免疫调节的作用机制,观察IL-2在脾脏中的具体分布特点、IL-2在脾脏中随年龄增长而发生的可能性变化,为以后研究IL-2在鹌鹑体内如何发挥免疫效应提供理论基础。随机选0,1,2,3,4,5,6周龄的健康黄羽鹌鹑各10只,取其脾脏,运用免疫组织化学法对IL-2进行组织学定位,观察不同周龄脾脏中阳性细胞的形态及分布情况,运用数据分析软件对所得数据进行分析,探寻IL-2随周龄增长是否存在变化规律。结果显示,阳性细胞主要出现在动脉周围淋巴鞘附近,部分在椭球周围淋巴鞘聚集,其余散在分布于脾索中;在0~6周龄中,IL-2的表达随着周龄增强,阳性细胞数量变化清晰可见,整体呈上升趋势,具有规律性。Tfh细胞和B淋巴细胞在脾脏发挥免疫功能时具有不同的作用,本试验中IL-2可能表达于迁移到PELS附近的Tfh细胞,也可能在PELS附近的B淋巴细胞中表达,表明IL-2阳性细胞分布与IL-2对淋巴细胞的作用密切相关;脾脏结构和功能随年龄的增长而发育,IL-2阳性细胞数量和免疫强度也随周龄的增长而呈上升趋势,表明IL-2阳性细胞免疫强度与脾脏组织结构和功能完善程度有关;以上说明IL-2可能参与脾脏T、B细胞的增殖分化,对脾脏免疫功能日益完善有重要作用。In order to explore the mechanism of IL-2 immunoregulation in the spleen of Chinese yellow feather quail,observe the specific distribution characteristics of IL-2 in the spleen and the possible changes of IL-2 in the spleen with the age,so as to provide a theoretical basis for the future study of how IL-2 plays a role in the immune response of quail.Ten healthy yellow feathered quails of 0,1,2,3,4,5 and 6 weeks old were randomly selected.Then the spleen of quails was taken and the IL-2 was histologically located by immunohistochemistry.The morphology and distribution of the positive cells in the spleen of different weeks old were observed.The data were analyzed by data analysis software to find out whether the IL-2 changed with the increase of weeks old.The results showed that the positive cells mainly appeared near the peri-arterial lymphoid sheath,some gathered in the peri-ellipsoidal lymphatic sheath,and the others were scattered in the splenic cord,and the expression of IL-2 increased with the age of 6 weeks,and the change of the number of positive cells was clear,showing an upward trend as a whole,with regularity.Tfh cells and B lymphocytes played different roles in the immune function of spleen.IL-2 might be expressed in Tfh cells migrated to PELS or in B lymphocytes near PELS,indicating that the distribution of IL-2 positive cells was closely related to the effect of IL-2 on lymphocytes.The structure and function of spleen developed with age,and the number and immune intensity of IL-2 positive cells increased with age,which indicated that the immune intensity of IL-2 positive cells was related to the perfection of spleen tissue structure and function;These results suggested that IL-2 might be involved in the proliferation and differentiation of spleen T cells and B cells and played an important role in the improvement of spleen immune function.
