摘  要:目的探讨天麻素对焦虑样胃高敏感性功能性消化不良(FD)模型大鼠脑胃调节因子的影响。方法将40只1日龄SPF级雄性SD仔鼠随机分为对照组、模型组、丁螺环酮组、天麻素(Gas)小剂量组和大剂量组,每组8只。出生第2日起采用序贯应激法,即序贯给予胃刺激和母婴分离,成年后给予束缚应激而制作焦虑样胃高敏感FD大鼠模型。于第8周束缚应激试验结束后开始腹腔注射给予对照组和模型组生理盐水2.0 mL/kg,丁螺环酮组给予丁螺环酮3.125 mg/kg,Gas小、大剂量组分别给予Gas 62.5 mg/kg和125.0 mg/kg,每天1次,连续7天。末次给药后采静脉血,并立即断头处死取脑和胃,用于测定海马、血浆和胃组织中5羟色胺(5-HT)、γ氨基丁酸(GABA)、脑源性神经营养因子(BDNF)和nesfatin-1的含量。结果与对照组相比,模型组大鼠各部位5-HT、BDNF含量和海马内GABA含量均明显减少,而nesfatin-1含量明显增加(P<0.01)。Gas大、小剂量组和丁螺环酮组各部位5-HT、BDNF含量和海马内GABA含量均明显增加,而nesfatin-1含量明显减少,组间比较差异具有统计学意义(P<0.01)。结论Gas能明显增加序贯应激所致焦虑样胃高敏感FD模型大鼠海马、血浆和胃组织中的5-HT、BDNF、GABA的含量,减少nesfatin-1的含量。Objective To investigate the effects of gastrodin on the levels of neurobiochemical modulators shared by brain and stomach in a rat model of FD with anxiety-like gastric hypersensitivity induced by sequential stress.MethodsRats were sequentially subjected to acute gastric irritation(AGI)and maternal separation(MS)early in life and restraint stress(RS)in adulthood.Then they were given intraperitoneal injection of 0.9%saline(2.0 mL/kg,for control and model group),buspirone(3.125 mg/kg,for buspirone group)or Gastrodin(Gas,62.5 or 125.0 mg/kg for Gas low or high dose groups),respectively(one time a day at 8:00 o’clock a.m.for 7 consecutive days).After that blood collection were performed and the rats were finally sacrificed by decapitation.The hippocampus and gastric fundus were promptly isolated and stored at-80℃for ELISA measurements of 5-hydroxytryptamine(5-HT),γ-aminobutyric acid(GABA),brain-derived neurotrophic factor(BDNF)and nesfatin-1.Results Compared with the control group,the hippocampal,plasmic and gastric levels of 5-HT,BDNF and GABA decreased significantly(P<0.01),while the concentrations of nesfatin-1 increased greatly(P<0.01).Gas could significantly attenuate the diminution of the hippocampal,plasmic and gastric levels of 5-HT,BDNF and GABA,but inhibit the increase of nesfatin-1 in the same sites(P<0.01 vs.model rats).Conclusions The results implied that Gas could influence the levels of 5-HT,GABA,BDNF and nesfatin-1 in hippocampus,plasma and gastric tissue of the sequential-stress-treated rats,and could be used in the treatment of FD with anxiety-like gastric hypersensitivity.
