摘  要:目的通过网络药理学方法探究参附注射液防治心肌顿抑的作用机制。方法利用中药系统药理数据库和分析平台(TCMSP)数据库,筛选出参附注射液中的有效化合物,用PharmMapper服务器和Uniprot数据库对有效化合物的靶点进行预测并统一基因名;通过GeneCards、OMIM和CTD数据库筛选心肌顿抑的相关靶点,将疾病和化合物靶点上传到Venny2.1在线分析软件中,获得疾病化合物的交集靶点,将所得交集靶点上传到David在线分析软件中进行GO分析和KEGG富集分析。结果对结果进行筛选后,获得43个有效化合物,74个相应靶点。与心肌顿抑发病机制相关靶点交集有26个,包括CASP3、EGFR、MAPK1、MAPK8、ESR1、MAPK14、CASP7、GSTP1等。通过GO分析得出55个生物学过程,15个细胞组成,20个分子功能。KEGG分析得出相关通路12条,主要通过TNF、Foxo、NOD样受体、MAPK、Toll样受体等信号通路对心肌顿抑进行干预。结论本文初步探讨了参附注射液通过多靶点、多通路对心肌顿抑干预的作用机制,分析发现与抑制炎症反应、减少细胞凋亡、降低氧化应激、调节组织修复等密切相关。Objective:To explore the mechanism of Shenfu Injection in preventing and treating myocardial stunning by means of network pharmacology.Methods:The Chinese Medicine System Pharmacological Database and Analysis Platform(TCMSP)database was used to screen out the effective compounds in Shenfu Injection were,and the PharmMapper server and Uniprot database were used to predict the effective compound targets and unify the gene names;through GeneCards,OMIM and The CTD database,the relevant targets of myocardial stunning were screened out;the disease and compound targets were uploaded to the Venny2.1 online analysis software to obtain the intersection target of the disease compound,and the obtained intersection target was uploaded to the David online analysis software for GO analysis and KEGG enrichment analysis.Results:After screening the results,43 effective compounds and 74 corresponding targets were obtained.There were 26 targets that were related to the pathogenesis of myocardial stunning,including CASP3,EGFR,MAPK1,MAPK8,ESR1,MAPK14,CASP7,GSTP1 and so on.Through GO analysis,55 biological processes,15 cell components,and 20 molecular functions were obtained.KEGG analysis revealed 12 related pathways,which intervened in myocardial stunning mainly through TNF,Foxo,NOD-like receptor,MAPK,Toll-like receptor and other signaling pathways.Conclusion:This article preliminarily discussed the mechanism of Shenfu Injection on myocardial stunning through multiple targets and multiple pathways.The analyses show that it is closely related to inhibiting inflammation,reducing cell apoptosis,reducing oxidative stress,and regulating tissue repair.
