活血潜阳祛痰方对肥胖高血压大鼠心肌氧化应激及炎症的影响

摘　　要：目的观察活血潜阳祛痰方对肥胖高血压(OBH)大鼠心肌氧化应激及炎症的影响。方法自发性高血压大鼠(SHR)喂养高脂饮食制备OBH大鼠,随机分为OBH组、中药低剂量组、中药高剂量组和缬沙坦组。以Wistar大鼠(WKY)和SHR为对照。治疗10周后测量体质量、左心室质量,观察心肌病理,检测心脏超声、血清和心肌中血管紧张素Ⅱ(AngⅡ)水平、心肌中丙二醛(MDA)、锰超氧化物歧化酶(Mn-SOD)、NADPH氧化酶4(NOX4)、核因子κB(NF-κB)、细胞外信号调节激酶1/2(ERK1/2)和p-ERK1/2的水平,对相关数据统计分析。结果与WKY组比较,OBH组左室质量指数(LVMI)和矫正的左心室质量(LVM-cor)均显著升高(P<0.05),血清和心肌AngⅡ水平明显升高(P<0.05),心肌中NOX4、NF-κB p65蛋白表达和ERK1/2磷酸化比值明显升高(P<0.05)。中药高剂量组的心脏质量指数、LVM-cor和血清AngⅡ水平较模型组降低(P<0.05)。中药各剂量组的LVMI、心肌AngⅡ水平和MDA值均较OBH组显著降低(P<0.05),心肌NOX4、NF-κB p65蛋白表达量和ERK1/2磷酸化比值也显著降低(P<0.05),Mn-SOD值高于OBH组(P<0.05)。结论活血潜阳祛痰方改善OBH心肌重构机制可能与抑制氧化应激,减轻炎症反应相关。Objective:To observe the effect of Huoxue Qianyang Qutan Decoction on myocardial oxidative stress and inflammation in obesity and hypertensive(OBH)rats.Methods:Spontaneously hypertensive rats(SHR)were fed with high-fat diet to prepare OBH rats,then they were randomly divided into the OBH group,traditional Chinese medicine low-dose group(ZD group),high-dose group(ZG group)and Valsartan group(X group).Kyoto Wistar rats(WKY)and SHR were used as controls.After 10 weeks of treatment,the body weight and left ventricular weight were measured,the pathological changes of myocardium were observed,the cardiac ultrasound was detected.The levels of MDA,Mn-SOD,NOX4,NF-κB,ERK1/2 and p-ERK1/2 in myocardium were detected.The related data were statistically analyzed.Results:Compared with WKY group,the left ventricular weight index(LVMI)and corrected left ventricular weight(LVM-cor)were significantly increased in OBH group(P<0.05),the AngⅡlevels in serum and myocardium were significantly increased(P<0.05),and the expression of NOX4,NF-κB p65 protein and ERK1/2 phosphorylation ratio were significantly increased(P<0.05).The heart weight index,LVM-cor and serum AngⅡlevel in ZG group were lower than OBH group(P<0.05).The LVMI,myocardial AngⅡlevel and MDA value in ZG group and ZD group were both significantly lower than those in OBH group(P<0.05),and myocardial NOX4 and NF-κB p65 protein expression,ERK1/2 phosphorylation ratio were significantly lower than those of OBH group,and Mn-SOD value was higher than that of OBH group(P<0.05).Conclusion:The mechanism of Huoxue Qianyang Qutan Decoction in improving myocardial remodeling of OBH may be related to the inhibition of oxidative stress and the reduction of inflammatory response.
