平喘颗粒通过抑制miR-21的表达对气道炎症及Eotaxin mRNA的影响

摘　　要：目的观察平喘颗粒通过抑制miR-21的表达对气道炎症及Eotaxin mRNA的影响。方法选择120只Wistar大鼠随机分为正常组、模型组、miR-21抑制表达组、miR-21抑制表达载体对照组、miR-21抑制表达+中药组、中药组。正常组给予生理盐水雾化,模型组、miR-21抑制表达组、miR-21抑制表达载体对照组、miR-21抑制表达+中药组、中药组应用卵蛋白吸入法制成慢性哮喘动物模型。miR-21抑制表达组、miR-21抑制表达载体对照组、miR-21抑制表达+中药组、中药组在造模前给予miR-21慢病毒溶液滴鼻造模。在末次激发24 h后观察大鼠的一般状态、BALF及外周血中EOS计数、用图像分析方法测定支气管基底膜厚度(WAi/Pi)和平滑肌层厚度(WAm/Pi)、Real-time PCR法检测miR-21、Eotaxin m RNA的表达。结果与空白组比较,模型组的BALF及外周血中EOS计数、WAi/Pi、WAm/Pi、miR-21、Eotaxin m RNA表达量显著升高(P<0.01);与模型组比较,miR-21抑制表达+中药组、中药组均可明显降低上述指标(P<0.05或P<0.01)。结论平喘颗粒能够通过抑制miR-21的表达,降低Eotaxin m RNA表达,从而影响EOS水平,减轻气道炎症,起到治疗作用。Objective:To observe the effect of Pingchuan Granules on airway inflammation and Eotaxin mRNA by inhibiting the expression of miR-21.Methods:A total of 120 Wistar rats were randomly divided into blank group(group A),model group(group B),miR-21 expression inhibition group(Group C),miR-21 expression vector control group(Group D),miR-21 expression inhibition+traditional Chinese medicine group(Group E),and Chinese medicine group(Group F).Group A was given normal saline atomization,groups B,C,D,E,F were given ovalbumin inhalation to establish chronic asthma animal model.Groups C,D,E and F were given miR-21 lentivirus solution before modeling.The general state,EOS count in BALF and peripheral blood were observed 24 hours after the last challenge.The bronchial basement membrane thickness(WAi/Pi)and smooth muscle layer thickness(WAM/Pi)were measured by image analysis.The expression of miR-21 and eotaxin m RNA was detected by Realtime PCR.Results:Compared with the blank group,the EOS count,WAi/Pi,WAM/Pi,miR-21,Eotaxin mRNA expression in BALF and peripheral blood of model group were significantly increased(P<0.01);compared with model group,miR-21 inhibition+TCM group and TCM group could significantly reduce the above indexes(P<0.05 or P<0.01).Conclusion:Pingchuan Granules can reduce the expression of eotaxin mRNA by inhibiting the expression of miR-21,thus affecting the level of EOS,reducing airway inflammation and playing a therapeutic role.
