摘 要:目的探讨2种狼疮性肾炎(lupus nephritis,LN)小鼠的高迁移率族蛋白-1(high mobility group box-1,HMGB-1)表达情况。方法选取MRL/lpr雄性小鼠(红斑狼疮肾炎小鼠)26只作为观察1组,取雄性C57BL/6小鼠24只作为对照1组,选择BXSB雄性小鼠(红斑狼疮肾炎小鼠)25只作为观察2组,野生型小鼠25只作为对照2组。采用电镜和PAS染色观察4组小鼠的肾组织结构改变,采用免疫组化法检测4组小鼠肾组织中HMGB-1的表达,采用酶联免疫吸附法检测4组小鼠血清中HMGB1的表达。结果电镜下观察1和2组有典型狼疮性肾炎病理表现,对照1和2组的肾组织结构正常;免疫组化法检测:观察1和2组小鼠肾组织的HMGB1表达高于对照1和2组(P<0.05);酶联免疫吸附法检测观察1和2组血清HMGB1表达水平显著高于对照1和2组(P<0.05)。结论 HMGB-1可能通过致炎作用参与了狼疮性肾炎的发病过程。Objective The expression of high mobility group box-1(HMGB-1) in 2 kinds of lupus nephritis mice. Methods Twenty-six MRL/lpr male mice(lupus nephritis mice) were selected as the observation group 1 and 24 male C57 BL/6 mice were used as the control group 1. 25 BXSB mice(lupus nephritis mice) were selected as the observation group 2, and 25 wild type mice were used as the control group 2. The renal tissue structure of 4 groups was observed by electron microscope and PAS staining. The expression of HMGB-1 in the kidneys of 4 groups was detected by immunohistochemistry. The HMGB-1 in the serum of 4 groups was detected by enzyme-linked immunosorbent assay(ELISA). Results The pathological manifestations of typical lupus nephritis in the observation group 1 and group 2 were found under electron microscope. The renal tissue structure of the control group 1 and group 2 was normal. Immunohistochemical method showed that the expression of HMGB-1 in kidney tissues of the observation group 1 and 2 was higher than that of the control group 1 and 2(P<0.05). ELISA showed that the expression of HMGB-1 in the observation group 1 and group 2 was significantly higher than that in the control group 1 and 2(P<0.05). Conclusion HMGB-1 may participate in the pathogenesis of lupus nephritis through inflammation.
