摘  要:目的:研究人参茎叶总皂苷的人肠内细菌生物转化。方法:采用液相色谱-电喷雾离子源-四极杆-飞行时间质谱法(LC-ESI-Q-TOF-MS)负离子监测模式,对人参茎叶总皂苷样品和人肠内细菌转化的人参茎叶总皂苷样品进行定性分析,将采集的原始数据导入分析软件进行碎片和相对分子质量匹配,根据高分辨质谱信息推导可能的裂解碎片;部分化合物通过与对照品比对进行解析,比较2个样品中人参达玛烷三萜化合物的差别。结果:在人参茎叶总皂苷样品中共鉴定出83个化合物,在人肠内细菌转化人参茎叶总皂苷的样品中共鉴定出67个化合物,包括区别性化合物20(S)-25-OH-原人参三醇[20(S)-25-OH-PPT]、20(R)-25-OH-PPT、人参皂苷O(G-O)、绞股蓝皂苷Ⅸ、绞股蓝皂苷ⅩⅦ、25-OH-G-Rh_(2)、20(22)E-G-Rg_(9)、G-Rs_(3)、20(R)-G-Rs_(3)、G-Mc、G-Y、G-Mx、20(S)-25-OH-原人参二醇[20(S)-25-OH-PPD]、G-CK和G-CK异构体。结论:人参茎叶总皂苷经人肠内细菌生物转化后产生了低极性的达玛烷型人参三萜类化合物;提示人参茎叶总皂苷经人肠内细菌生物转化后更利于其吸收进入系统循环。Objective:To study the biotransformation of total saponins from the stems-leaves of Panax ginseng(GTSSL)with human intestinal flora.Methods:The samples of GTSSL and its biotransformation products with human intestinal flora were qualitatively analyted by liquid chromatography-electrospray ionization-quadrupole-time-of-flight-mass spectrometry(LC-ESI-Q-TOF-MS)in the negative ion mode,which the collected raw data were imported analysis software to match with fragmentation and molecular weight.Then,the molecular formula and their fragmentation were deduced according to the information of high-resolution mass spectrometry.And a part of compounds were determined by comparison with referent substances.Basis on the above-mentioned strategy,the differences of ginseng-dammarane-triterpenes in the two samples were compared.Results:A total of 83 compounds were identified in the GTSSL.While a total of 67 compounds were identified in the biotransformation products with human intestinal flora of GTSSL,including differentiative compounds 20(S)-25-OH-protopanaxatriol[20(S)-25-OH-PPT],20(R)-25-OH-PPT,G-O,gypenosideⅨ,gypenosideⅩⅦ,25-OH-G-Rh_(2),20(22)E-G-Rg_(9),G-Rs_(3),20(R)-G-Rs_(3),G-Mc,G-Y,G-Mx,20(S)-25-OH-protopanaxadiol[20(S)-25-OH-PPD],G-CK and G-CK isomer.Conclusion:The low polar ginseng-dammarane-triterpenes were produced in the biotransformation products with human intestinal flora of GTSSL.It was prompted that GTSSL was more easily absorbed into systemic circulation after the biotransformation by human intestinal flora.
