摘 要:骨性关节炎(osteoarthritis, OA)是以关节软骨退变为主要特征的退行性疾病,软骨细胞代谢失衡是导致其发生、发展的重要原因。转化生长因子-β(transforming growth factorβ, TGF-β)超家族作为生长因子在软骨细胞增殖、表型的维持、细胞外基质的合成中发挥着重要作用。但过度活化的TGF-β超家族却促进软骨细胞肥大、凋亡、软骨内骨化进而推动OA的发展。分泌磷蛋白24 (secreted phosphoprotein 24, Spp24)拥有与TGF-β超家族成员的高亲和性,能否作为OA的潜在治疗靶点值得思考。本文就TGF-β超家族在骨性关节炎软骨细胞退变中的角色及Spp24的潜在作用做一综述。Osteoarthritis is a degenerative disease characterized by degeneration of articular cartilage, which is related to the imbalance of chondrocyte metabolism. Transforming growth factor β(TGF-β) superfamily, as a growth factor, plays an important role in chondrocyte proliferation, maintenance of phenotype and synthesis of extracellular matrix. However, overactivated TGF-β superfamily promotes chondrocyte hypertrophy, apoptosis, and endochondral bone growth to promote the development of osteoarthritis. In addition, secreted phosphoprotein 24(Spp24), with high affinity for TGF-β superfamily members, is worth considering as a potential therapeutic target for osteoarthritis. This article reviews the role of TGF-β superfamily in chondrogenic degeneration of osteoarthritis and the potential role of Spp24.
