摘 要:目的研究酒石酸唑吡坦在模拟失重与正常SD大鼠体内十二指肠的吸收特性,并比较酒石酸唑吡坦在正常和模拟失重大鼠中十二指肠吸收变化。方法采用大鼠在体单向肠灌流模型,利用高效液相色谱(HPLC)法测定肠灌流液中酒石酸唑吡坦的含量,计算有效渗透系数(P_(eff))和吸收速率参数(K_(a))。结果酒石酸唑吡坦在模拟失重SD大鼠体内十二指肠段的吸收参数与正常SD大鼠吸收参数的差异有统计学意义(P<0.05),其中模拟失重状态下大鼠十二指肠P_(eff)和K_(a)均减小。结论酒石酸唑吡坦在模拟失重状态下SD大鼠体内十二指肠P_(eff)和K_(a)均小于正常组SD大鼠,因此模拟失重状态会减少酒石酸唑吡坦在SD大鼠十二指肠段吸收程度和吸收速度。提示机体进入失重环境后需要调整酒石酸唑吡坦服用剂量。Objective To investigate the duodenum absorption characteristics of zolpidem tartrate and explore the changes between normal SD rats and simulated weightlessness SD rats.Methods The concentrations of zolpidem tartate in single pass intestinal perfusion(SPIP)model were determined by HPLC.The effective permeability coefficient(P_(eff))and absorption rate parameter(K_(a))were calculated.Results The duodenum absorption characteristics of zolpidem tartrate in simulated weightlessness SD rats and normal rats had a significant difference(P<0.05).The P_(eff)and K_(a)of duodenum were decreased in simulated weightlessness groups.Conclusion The P_(eff)and K_(a)of duodenum in simulated weightlessness SD rats were lower than those in normal rats.The simulated weightlessness had a great influence on the absorption of zolpidem tartrate in the duodenum of SD rats.These results suggest that it is necessary to adjust the dosage of zolpidem tartrate after entering the weightlessness environment.
