摘 要:目的研究冬虫夏草提取液(CSE)对辐照诱导小鼠睾丸组织损伤的保护机制。方法50只雄性C57BL/6小鼠随机分为5组:对照(Control)组、辐照(Model)组、低剂量CSE治疗(CSE-L)组、中剂量CSE治疗(CSE-M)组、高剂量CSE治疗(CSE-H)组,每组各10只。除Control组外,其余各组均以单次8 Gy~(60)^Coγ射线全身辐照小鼠,建立睾丸损伤模型。CSE-L、CSE-M、CSE-H组小鼠生药剂量分别为312.5、625、1250 mg/kg,每日灌胃给药1次,持续给药28 d,检测不同生药剂量的CSE对辐照后小鼠体质量的影响,并对Control、Model组、CSE-H组小鼠的睾丸组织进行了TUNEL染色、凋亡相关基因及蛋白表达水平的检测。结果与Control组相比,Model组小鼠体质量显著降低(P<0.05),表明造模成功。与Model组相比,CSE治疗组均能显著改善辐照小鼠的体质量(P<0.05),且呈剂量依赖性。TUNEL结果显示:CSE-H组中辐照诱导的小鼠睾丸组织TUNEL阳性细胞数目显著降低。此外,与Control组相比,Model组中小鼠睾丸组织抗凋亡基因及蛋白Bcl-2表达水平明显降低;促凋亡基因(Bax、caspase 3)mRNA与蛋白(Bax、cleaved-caspase 3)表达量显著上升。与Model组相比,CSE-H组中小鼠睾丸组织抑凋亡基因与蛋白表达水平明显上调,促凋亡基因与蛋白水平显著下调。结论CSE对电离辐照导致的睾丸组织细胞损伤有保护作用。Objective To investigate the protective mechanism of Cordyceps sinensis extract(CSE)on the testicular tissue cells of mice exposed to irradiation.Methods Fifty C57BL/6 male mice were randomly divided into 5 groups:the control group(the non-irradiated group),the model group(the irradiation group),the CSE low dose group(the CSE-L group),the CSE medium dose group(the CSE-M group)and the CSE high dose group(the CSE-H group),each consisting of 10 animals.With the exception of the control group,all the other groups received total-body irradiation with a single dose of 8 Gy ^(60)Coγand the animal testicular tissue injury model was thus established.The animals in the CSE-L,CSE-M,and CSE-H groups all had CSE gavage once a day,at the dosages of 312.5 mg/kg,625 mg/kg and 1250 mg/kg respectively,for a succession of 28 days.Then,detections were made to identify the effects of different CSE doses on the body mass indexes of the mice following irradiation.Finally,TUNEL was performed in the testicular tissues of the control group,the model group and the CSE-H group,and detections on apoptosis-related genes and the expression levels were conducted as well.Results Compared with that of the control group,the body mass index of the model group markedly decreased,indicating success of model development(P<0.05).Compared with the model group,the body mass indexes of the CSE groups with various doses were improved in an obviously dose-dependent manner(P<0.05).TUNEL staining results showed that the number of TUNEL-positive cells in the CSE-H group dropped markedly.Besides,as compared with those of the control group,the expression levels of anti-apoptosis genes and Bcl-2 in the testicular tissues of the mice in the model group decreased dramatically,while the expression levels of the pro-apoptosis genes(Bax、caspase 3)and the proteins(Bax、cleaved-caspase 3)in the model group elevated markedly.In contrast to the model group,the anti-apoptosis genes and proteins levels in the testicular tissues of the CSE-H group were considerably up-re
