摘 要:目的探讨扶正消积汤含药血清通过下调miR-184促进人肺腺癌SPC-A1细胞凋亡的影响。方法将实验分为对照组、扶正消积汤含药0.05%、0.1%和0.2%组,分别处理24 h和48 h后对其进行检测,使用倒置显微镜观察各组细胞形态的变化,使用CCK-8法检测细胞增殖抑制率,使用流式细胞仪检测并计算细胞凋亡率,实时定量PCR检测PIEN和PDCD4 mRNA表达水平,最后使用Graphpad Prism 5软件进行统计分析。结果CPS-A1细胞的活力随着扶正消积汤含药浓度和时间的上升而下降,并且细胞增殖抑制率显著上升,当扶正消积汤含药为0.2%时,细胞增殖抑制率在48 h可达到80%;随着扶正消积汤含药浓度的上升,细胞形态可见明显变化,细胞在0.2%的浓度时,开始出现细胞膜边缘出现小气泡和整体萎缩等凋亡形态;SPC-A1细胞早期凋亡率随着扶正消积汤含药浓度的升高而升高,并且可见48 h的细胞早期凋亡率显著高于24 h;miR-184在0.1%浓度扶正消积汤含药血清处理SPC-A1细胞12 h后明显下降,PTEN和PDCD4表达则显著上升。结论扶正消积汤含药血清可显著抑制人肺腺癌细胞SPC-A1的细胞增殖,并可改变细胞的形态,抑癌基因PTEN和PDCD4的表达随着miR-184表达的下降而上升,从而达到促进肺癌细胞凋亡的抗肿瘤作用。Objective To investigate the effect of Fuzheng Xiaoji Decoction serum on the apoptosis of human lung adenocarcinoma SPC-A1 cells by down regulating mir-184.Methods The experiment was divided into control group,Fuzheng Xiaoji Decoction containing 0.05%,0.1%and 0.2%groups.After treatment for 24 and 48 hours,the changes of cell morphology were observed by inverted microscope,the inhibition rate of cell proliferation was detected by CCK-8 method.The apoptosis rate was detected and calculated by flow cytometer.The expression level of Pien and PDCD4 mRNA was detected by real-time quantitative PCR,and finally,graphph was used Ad prism 5 software for statistical analysis.Results The viability of CPS-A1 cells decreased with the increase of the concentration and time of Fuzheng Xiaoji Decoction,and the inhibition rate of cell proliferation increased significantly.When the content of Fuzheng Xiaoji Decoction was 0.2%,the cell proliferation inhibition rate could be reached within 48 hours.Reach 80%;with the increase of the concentration of Fuzheng Xiaoji Decoction,the cell morphology changes significantly.At 0.2%concentration,the cell membrane edge appears small bubbles and overall atrophy and other apoptotic forms;SPC-A1 cells in the early stage The apoptosis rate increases with the increase of the concentration of Fuzheng Xiaoji Decoction,and it can be seen that the early apoptosis rate of cells at 48h is significantly higher than that at 24 h;miR-184 is treated with Fuzhengxiaoji Decoction at a concentration of 0.1%SPC-A1 cells showed a significant decline after 12 h,while the expression of PTEN and PDCD4 increased significantly.Conclusion Fuzheng Xiaoji Decoction can significantly inhibit the proliferation of human lung adenocarcinoma cells SPC-A1 and change the cell morphology.The expression of tumor suppressor genes PTEN and PD-CD4 increases with the decrease of miR-184 expression.So as to achieve the anti-tumor effect of promoting lung cancer cell apoptosis.
