摘  要:目的观察戟天健脑方对老龄血管性痴呆(VD)模型大鼠海马炎症细胞因子IL-1β、TNF-α的影响。方法70只老龄SD大鼠随机分为空白组、假手术组、模型组、尼莫地平组,戟天健脑方低、中、高剂量组,每组10只。采用双侧颈总动脉永久性结扎法(2-VO法)制备成VD大鼠模型。戟天健脑方低、中、高剂量组分别给予戟天健脑方5.4 mg/(kg·d)、10.8 mg/(kg·d)、21.6 mg/(kg·d)灌胃,尼莫地平组给予尼莫地平50 mg/(kg·d)灌胃,空白组、模型组和假手术组分别给予等量生理盐水灌胃,均2次/d。各组大鼠均连续灌胃4周,然后进行Morris水迷宫实验,并观察海马CA1区神经元组织形态的变化和测定海马组织IL-1β和TNF-α的含量。结果戟天健脑方高、中剂量组大鼠穿台次数、目标象限活动路程较模型组明显增加(P<0.05),戟天健脑方高剂量组大鼠目标象限活动时间明显增加(P<0.05);戟天健脑方高、中剂量组及尼莫地平组海马锥体细胞较模型组细胞数量增多,排列较整齐,细胞间隙减小;与模型组比较,尼莫地平组TNF-α蛋白含量明显降低(P<0.05),戟天健脑方高剂量组可明显降低IL-1β含量(P<0.05),戟天健脑方中、高剂量组均可明显降低TNF-α含量(P<0.05)。结论戟天健脑方能够改善VD大鼠学习记忆能力,减轻VD大鼠海马CA1区的损伤,下调海马组织炎症因子IL-1β和TNF-α的表达,发挥保护神经元的作用。Objective To observe the effect of Jitian Jiannao Decoction on hippocampal inflammatory cytokines IL-1βand TNF-αin aged rats with vascular dementia.Methods Seventy old SD rats were randomly divided into blank group,sham operation group,model group,nimodipine group,small,medium and high dose groups of Jitian Jiannao Decoction,with 10 rats in each group.The VD rat model was prepared by permanent ligation of bilateral common carotid arteries(2-VO method).The small,medium and high-dose groups of Jitian Jiannao Decoction were given 5.4 mg/(kgod),10.8 mg/(kgod),21.6 mg/(kgod)by gavage,Nimo The dipine group was given nimodipine 50 mg/(kgod)by gavage,and the blank control group,model group and sham operation group were given the same amount of normal saline gavage,all twice a day.The rats in each group were given intragastric administration for 4 weeks,and then the Morris water maze experiment was performed to observe the changes of neuronal tissue morphology in the hippocampal CA1 area and measure the levels of IL-1βand TNF-αin the hippocampus.Results he number of times of traversing the table and the distance of movement in the target quadrant of the rats in the high and medium doses of Jitian Jiannao Decoction were significantly increased compared with the model group(P<0.05),and the time of the rats in the high dose group of Jitian Jiannao Decoction was significantly increased(P<0.05);Compared with the model group,the number of pyramidal cells in the hippocampus of the high-dose and medium-dose groups of Jitian Jiannao Decoction and the nimodipine group were increased,the arrangement was more orderly,and the intercellular space was reduced;compared with the model group,nimodipine The TNF-αprotein content in the group was significantly reduced(P<0.05),the high-dose Jitian Jiannao Decoction group could significantly reduce the IL-1βcontent(P<0.05),and the middle and high-dose Jitian Jiannao Decoction groups could significantly reduce TNF-αprotein content(P<0.05).Conclusion Jitian Jiannao Decoction can improve th
