摘 要:热激蛋白Hsp90是广泛存在于微生物和动植物体中高度保守蛋白质之一,大量研究发现其参与生物体应答应激反应、信号转导、细胞周期调控、蛋白降解及转运等生理活动,具有重要研究价值,因此在许多物种中热激蛋白Hsp90基因家均被广泛关注和研究。但大豆中Hsp90基因家族详细信息尚不清楚,研究通过生物信息学分析发现共有13个Hsp90基因在大豆中被鉴定出来,基因序列长度2538~9131 bp,编码序列长度1968~2533 bp,外显子数为2~19个,编码蛋白质氨基酸个数为655~847,其分子质量为75.88~97.07 ku,理论等电点为4.84~6.28,氨基酸序列高度保守,同源性为62%,均包含Hsp90结构域pfam00183和HATPase_C结构域pfam02518,主要分布于8条染色体;系统进化树分析结果显示大豆Hsp90家族基因分为3组;亚细胞定位预测结果显示大豆Hsp90家族基因主要位于细胞质和内质网。研究结果为进一步探索大豆Hsp90家族成员功能奠定了基础。Heat shock protein Hsp90 is one of the highly conserved proteins widely found in microorganisms and animals and plants,a growing number of studies have found that it involve in biological response to stress reaction,signal transduction,cell cycle regulation,protein degradation and transportand other physiological activities.It has important research value,so Hsp90 gene families has been studied extensively in many species.Soybean Hsp90 gene family details are unclear now,in this study,a total of 13 Hsp90 genes were identified through bioinformatics analysis in soybean,gene sequence length was 2538-9131 bp,coding sequence length was 1968-2533 bp,the number of exon was 2-19,the number of amino acid which coded protein was between 655-847,and its molecular weight was 75.88-97.07 ku,the theory of isoelectric point was 4.84-6.28,the amino acid sequence was highly conserved,its homology of 62%,both contained the Hsp90 domain PFAM00183 and Hatpase C domain PFAM02518,the Hsp90 genes were mainly distributed on eight chromosomes.Phylogenetic tree analysis showed that the Hsp90 family genes of soybean were divided into three groups.The prediction results of subcellular localization showed that the Hsp90 family genes were mainly located in cytoplasm and endoplasmic reticulum.The results of this study laid a foundation for further study on the function of soybean Hsp90 family members.
