摘 要:目的观察龟羚帕安丸对帕金森病(Parkinson’s disease,PD)大鼠模型中脑黑质神经细胞病理及自噬相关蛋白Beclin1、P62、LC3Ⅱ表达的影响。方法采用6-羟基多巴(6-OHDA)注射法建立PD大鼠模型,造模成功大鼠随机分为模型对照组、美多芭组、龟羚帕安丸高、中、低剂量组及假手术对照组,每组10只。对照组、模型组给予等容积生理盐水,其余组给予相应药物灌胃,连续给药30 d。观察各组大鼠中脑黑质病理改变;免疫组化法检测大鼠中脑黑质Beclin1、P62及LC3Ⅱ蛋白的表达变化情况。结果模型组神经元细胞出现较为明显的损伤;模型组中脑黑质Beclin1及LC3Ⅱ表达降低,P62表达明显升高;各治疗组大鼠神经元损伤情况较模型组有所改善,中脑黑质Beclin1及LC3Ⅱ表达明显升高,P62表达明显降低。结论龟羚帕安丸可明显改善神经细胞损伤,作用机制与提高Beclin1及LC3Ⅱ表达,降低P62的蓄积,疏通自噬-溶酶体降解途径,提高细胞自噬活性,改善细胞自身稳态有关。Objective To observe the effect of Guiling Paan Pill(龟羚帕安丸)on pathology and expressions of autophagy-related protein Beclin1,P62 and LC3Ⅱin substantia nigra neurons of Parkinson’s disease(PD)rats.Methods PD rat models were established by 6-hydroxydopa(6-OHDA)injection.The rats were randomly divided into model group,Madopar group,Guiling Paan Pill high,middle and low dose groups and control group(10 rats in each group).The control group and the model group were given the same volume of normal saline,and the other groups were given the corresponding drugs by gavage for 30 days.The changes of Beclin1,P62 and LC3Ⅱprotein expressions in the substantia nigra were observed by immunohistochemistry.Results The neurons of the model group were damaged obviously.The expressions of Beclin1 and LC3Ⅱin substantia nigra of the model group were decreased.The expression of P62 was increased obviously and the injury of neurons in the treatment group was improved compared with that of the model group.The expressions of Beclin1 and LC3Ⅱin substantia nigra of mesencephalon increased obviously,while the expression of P62 decreased obviously.Conclusion Guiling Paan Pill can obviously improve the injury of nerve cells.The mechanism is related to increasing the expressions of Beclin1 and LC3Ⅱ,decreasing the accumulation of P62,clearing the pathway of autophagy-lysosome degradation and improving the autophagy activity and the cell homeostasis.
