摘  要:目的探究覆盆子鞣质对大鼠肝微粒体内蛋白含量以及细胞色素p450(CYP450)和细胞色素b5(CYPb5)活性的影响。方法将30只SD雄性大鼠置于SPF环境中喂养1周后,随机分为5组,每组6只,分别为覆盆子鞣质高、中、低剂量组,阳性对照组和空白对照组。覆盆子鞣质高、中、低剂量组分别按150、100、75 mg·kg^(-1)·d^(-1)灌胃给药,空白对照组给予等量的生理盐水,连续灌胃给药7 d;苯巴比妥钠对照组连续5 d以80 mg·kg^(-1)·d^(-1)剂量经腹腔注射方式给药。结束给药后禁食12 h,利用一氧化碳(CO)还原差示光谱法测量各组肝微粒体中CYP450和CYPb5的含量。结果与空白对照组比较,覆盆子鞣质高、中、低剂量组均能降低CYP450和CYPb5含量,差异有统计学意义(P<0.01),且随着给药剂量的增加,抑制作用也随之增强。结论覆盆子鞣质对大鼠肝微粒体代谢酶CYP450和CYPb5的活性有显著的抑制作用,且呈现剂量依耐性。该结果为覆盆子鞣质的深度开发及临床应用等提供重要的实验依据。Objective To study the effects of tannins from Fupenzi(Rubus chingii Hu)on the protein in the hepatic microsomal of rats and the activated of cytochrome P450 and cytochrome b5.Methods Thirty male SD rats were randomly divided into five groups with 6 rats in each after feeding in the SPF environment for 7 d,which included high,medium and low dose groups of tannins from Fupenzi(Rubus chingii Hu),the positive control group and the blank control group.The three different doses groups were respectively given tannins from Fupenzi(Rubus chingii Hu)orally at the dose of 150,100 and 75 mg·kg^(-1)·d^(-1) and the blank control group was given physiological saline for 7 days.The positive control group was given phenobarbital sodium 80 mg·kg^(-1)·d^(-1) with intramuscular injection for 5 days.At the end of the experiment,the contents of cytochrome P450 and cytochrome b5 of liver microsomal in each group would be measured by CO reduction differential spectroscopy.Results Compared with the blank control group,the high,medium and low doses of tannins from Fupenzi(Rubus chingii Hu)could significantly inhibit the expressions of cytochrome P450 and cytochrome b5 enzymes(P<0.01).And with the increase of dosage,the inhibitory effect also increased.Conclusion There is a significant inhibitory effect on the activated of cytochrome P450 and cytochrome b5 in a dose-dependent manner when the rats were fed with tannins from Fupenzi(Rubus chingii Hu).The results provided an important experimental basis for further development and clinical application of tannins from Fupenzi(Rubus chingii Hu).
